Experiment of Meselsohn and Stahl


Based on Watson and Cricks double helix DNA model, three models of replication were proposed a) Conservative model- parental double helix remains and a second copy is made. b) Semi-conservative model- parental strands separate and each strand become a template for the synthesis of each new complementary strand. c) Dispersive model- both daughter strands have a mixture of the parent and the newly synthesized part.

Meselhon and Stahl (American Biochemist) proved that replication of DNA is semiconservative. Hybrid of DNA exists in every generation. In conservative model, there will be no hybrid of DNA formed. (Therefore, this model is NOT ACCEPTED) In dispersive model, the second generation will have no 14N14N DNA. (Therefore, this model is NOT ACCEPTED) The Semi-conservative model was accepted as the correct model for DNA replication in cell.
