What difficulties did you encounter in the preparation of your slides? I have no difficulties encountered in the preparation of my slides because the protocol was very to do and I observed lots of Anabaena instantly on my slide clearly which was the primary purpose of this exercise.
In this specific technique, which is better, a thin or thick suspension of algal filaments or colonies? A thin suspension of algal filaments or colonies is much better than a thick suspension. Because in a thin suspension, you can observed clearly the details and structures of the alga. Unlike the thick suspension, in which the colonies are overlapping to each other that that makes the algal structure be seen unclearly.
Did you observed discoloration of specimen? Yes I observed a yellow discoloration of the specimen.
