***Sanger Method -Need DNA in single stranded form (Template DNA) -Need DNA polymerase (to initiate DNA synthesis) -dNTPs -Single Primer
1. Add mixture of the above materials into four tubes each with a unique ddNTPs (dideoxyribonucleotide) thats fluorescently stained 2. Replication continues w/ dNTPs base pairing 3. Synthesis of DNA terminated when ddNTPs pairs with template DNA Result in each tube: multiple DNA strands
