ARRDC3 promotes the degradation of p27, a cell cycle inhibitor, by interacting with β-arrestin and targeting p27 for ubiquitination and proteasomal degradation. ARRDC3 recruits the deubiquitinating enzyme SIRT2, which removes ubiquitin from ARRDC3, allowing it to continuously transport p27 to the proteasome for degradation and promote cell cycle progression. Overexpression of ARRDC3 promotes cell proliferation by degrading p27.
