METHOD
Throat Swab Specimen:
1. Prepare the glass slide. Mark the area you are going to rub the specimen on, using a
marker at the other side of the glass (the side youre not going to rub the specimen on)
2. Prepare the swab.
3. Stand in front of your partner.
4. Instruct your partner to open his mouth as wide as possible, and make him say
Ahhhhhh
5. Direct the tip toward the tonsil area. DO NOT touch the swab tip to any other area of the
mouth, including the tongue.
6. Touch the swab tip to the tonsil area.
7. Rub the swab tip quickly and firmly over this area to obtain a good sample.
8. Remove swab from mouth (without touching any surface).
9. Rub the swab tip on the glass slide (only at the area that is already marked)
10. Fixate the glass slide with Bunsen.
Gram Staining:

Cover smear with Crystal Violet for 30 seconds

Wash off gently
Cover with Iodine for 10 seconds
Wash off gently
Decolorize with Ethyl Alcohol
Wash off gently
Cover with Safranin for 30 seconds
Wash off gently
Let it dry

METHOD
Teeth Swab Specimen:
1. Prepare the glass slide. Mark the area you are going to rub the specimen on, using a
marker at the other side of the glass (the side youre not going to rub the specimen on)
2. Cover area for specimen on the glass slide with NaCl.
3. Prepare the toothpick.
4. Open your mouth and grin.
5. Direct the tip of toothpick toward teeth.
6. Poke the tip of toothpick to area between teeth.
7. Remove toothpick from teeth.
8. Rub the tip of toothpick on the glass slide (only at the area that is already marked)
9. Fixate the glass slide with Bunsen.
Gram Staining:

Cover smear with Crystal Violet for 30 seconds

Wash off gently
Cover with Iodine for 10 seconds
Wash off gently
Decolorize with Ethyl Alcohol
Wash off gently
Cover with Safranin for 30 seconds
Wash off gently
Let it dry
