Lab 3: Mutagenesis of Yeast Cells with UV Light
1. Label 4 sterile Eppendorf Tubes: 10-1, 10-2, 10-3, and 10-4.
2. Put .9 ml of sterile water into each tube.
3. Put .1ml of yeast solution into tube 1 and mix
4. Take .1ml from tube 1 and put in tube 2 and mix
5. Put .1ml from tube 2 into tube 3 and .1ml of tube 3 into tube 4.
6. Obtain 7 SC Petri dishes and 7 SD-trp Petri dishes.
7. Put .1ml of tube 4 into each of the 7 SC dishes.
8. Heat the spreader and then spread the yeast throughout the dish.
9. Add .1ml of the original yeast solution to the 7 SD dishes and spread.
10. Label one plate from each with 0 and put aside.
11. Expose one plate from each set to 20, 40, 60, 80, 100, and 120 minutes.
12. Make sure to remove the plates and put on Saran Wrap while under UV light.
