– Consider the nocturnal periodicity of the filarial parasite to select the optimal
blood time (10 PM to 2 PM)
– Uses 20 uL of blood spread in the slide & stained with Giemsa
– Wet smear can also be made by diluting 20–40 uL of anti-coagulated blood with
water or 2% saponin which will lyse the red cells.

– Basis of the test: a polyclonal antibody (pAb) and a monoclonal antibody (mAb),
both specific for W. bancrofti
– Mounted on a credit card–sized cardboard
– Detects lymphatic antigens in infected humans
– When the card is folded onto itself, the sample and the pAb will come in contact
with the membrane containing the mAb
– Two lines = + result
– One line = negative
