The Line Probe Assay (LIPA)
y Initially developed at Innogenetics
y Easy and fast
y Determination of multiple genetic variations
y Detection and Quantification of Polymorphism
y This assays have recently been developed for
y HLA class I A, B, and C typing
y HIV reverse transcriptase drug resistance
y Determination of HPV polymorphism
y Determination of HBV genotypes and pre‐core mutants

Preparation of probes for LiPA
1. Employs the distinction of polymorphisms
specific for the different genotypes of an
organism
1. Probes of 15 ‐ 20 nucleotides were tailed with a
poly(T)‐tail by terminal deoxynucleotidyl
transferase
2. Attached to nitrocellulose membrane

Isolation of DNA or RNA and PCR
y The biotin group can be incorporated by addition of
y Biotinylated dUTP
y 5’‐biotinylated primer during PCR
y Biotinylated amplicons can be readily transferred to the LiPA
procedure.

BCIP / NBT / Alkaline phosphate system for
detection of reverse hybridization
y Combination of following compounds yield an

intense, insoluble black‐purple precipitate when
reacted with Alkaline Phosphatase
y BCIP (5‐Bromo‐4‐Chloro‐3'‐Indolyphosphate p‐

BCIP / NBT / Alkaline phosphate system for
detection of reverse hybridization
y BCIP is hydrolyzed by alkaline phosphatase to
form an intermediate that undergoes dimerization
to produce an indigo dye
BCIP / NBT / Alkaline phosphate system for
detection of reverse hybridization
y The NBT is reduced to the NBT
The NBT is reduced to the NBT‐formazan
formazan by the
two reducing equivalents generated by the
dimerization.
