Chairside Test for Periodontal Risk The BANA Test A Inexpensive, 5-minute, Chairside Periodontal Test Periodontal Risk Assessment Patient Compliance (Home care) Patient Compliance (Antibiotics). The BANA Test is the chairside version of the same enzyme test used in clinical studies to detect P. gingivalis, B. forsythus and T. denticola, the three most common and riskiest pathogenic bacteria, also known as the so-called Red Group. The test is just as sensitive as more expensive DNA tests, being both highly sensitive and highly specific. It can detect as few as 100 pathogenic microbes among the millions of bacteria in a typical pocket. Despite its amazing accuracy, the BANA test is simple to use, just lightly smear subgingival plaque on the test strip. Its also: inexpensive (just $6 per test); fast (5 minutes total test time in a fully automated, chairside processor); and easy to read (positive = blue color, negative = no color change). No complicated procedures. No exorbitant costs. No waiting. No kidding! How it works Using a curette, obtain biofilms from the apical third of as many sites as you like. Lightly smear the plaque onto the bottom pad on the BANA test strip. Moisten the upper pad with a bit of distilled water. Fold at the perforation and insert into one of the two independent slots in the processor. That automatically activates the processor. Five minutes later, a signal light and chime let you know when the cycle is complete. If P. gingivalis, B. forsythus or T. denticola were present in the sample, the test strip turns blue. The darker the color, the higher the concentration. Blood, saliva or other microbes have no effect on the accuracy of the test. of Michigan School of Dentistry. The test exploits a unique property common to only 3 oral species (Treponema denticola, Porphyromonas gingivalis and Bacteroides forsythias), the three bacteria most highly associated with adult periodontitis. These three microbes possess a an uncommon enzyme that they use to break down short segments of amino acids called peptides. The BANA test contains a synthetic peptide (Benzoyl-DL-Arginine NaphthylAmide or BANA for short) that reacts with the bacterial enzyme. If any of the three pathogens are present, the enzyme and peptide react to form B-naphthylamide, which, in turn, reacts with an embedded diazo dye in the test strip which then turns blue. The depth of the color is proportional to the number of pathogens and periodontal risk. For More Information Visit the BANA research website.
