Gilsons revolutionary Fraction Trapping System is unlike any other system on the market today. The Fraction Trapping System is ideally suited for processing fractions up to 20 mL in volume and concentrations up to 500 mg that have been purified on virtually any preparative HPLC system. Washing steps can be performed for removal of salts, buffers and mobile phase modifiers such as TFA that can hinder further LC/MS analysis. Fractions containing greater than even 50% water content can be processed and collected in 100% of organic solvent. This allows for fast dry down times at lower temperatures for samples that are heat sensitive. Dry down times of 12 24 hours can be reduced to less than 30 minutes.
Process fractions in 8 to 12 minutes with volumes up to 20 mL and concentrations up to 500 mg.

Optimizing Preparative HPLC

There are numerous parameters and techniques to optimize preparative HPLC. These reference tables provide important physical parameters of the tubing and injection valve structure; optimization will improve chromatography and percent recovery of collected fractions. Table 1: 33X Series pump tubing (PEEK)* At high flow rates tubing can cause high back pressure. Ensuring that the appropriate tubing ID/flow rate combination is used will optimize the pumping system.

Novel Approach for Screening of Drug Absorption via an Automated System The system presented provides a faster alternative to other screening methods, as well as an inexpensive analysis estimate of drug permeability for hundreds of drug candidates in a fraction of the time of conventional screening methods. Preparative HPLC: Factors and Parameters that Directly Affect Recovery of Collected Fractions Parameters discussed are column performance, mobile phase constituents, detector settings and collection parameters such as collection via level versus slope and delay volumes. The data offers guidelines for all chromatographers purifying compounds by preparative HPLC and can be implemented into existing systems. Optimized Sample Load Capacity and Separation Through a Series of Short Preparative Columns By placing the smaller preparative columns in series, loading capacity and separations of compounds can increase with short run times and semi-preparative flow rates, minimizing the overall cost of the larger preparative columns and channeling within the column. Train Keep a Rolling and Isolation Gradients: A Method for Purifying a Large Quantity of Sample via Continuous Injection and Simultaneous Fraction Collection on Multiple FC Beds Train injection employs continuous injection of sample under isocratic conditions and continuous fraction collection. In addition, isolation gradients also offer an efficient purification of preparative samples when the train injection technique is not applicable, as is the case with complicated chromatograms. Separation of Flavonoids Through Automated Multiple Column Injections and Sample Concentration via C18 Column Capture Identical columns are configured in parallel for multiple injections sequenced by the sample peak elution. Sample is concentrated on a separate C18 preparative column, the purified compound is eluted in high organic solvent for ease of dry-down. Enhanced Recovery of Compound Mixtures by Concurrent Dissection and Concentration of Isolates A crude separation of a complex mixture into impure isolates. The impure isolates are reintroduced to the column. Front and back sections are cleaved to fractionate uncontaminated compound on separate columns for concentration. Final elution dispenses pure compound in high organic solvent for ease of dry-down.

Automated Post-collection Concentration for Purified Preparative Fractions via SPE The method (as opposed to analytical applications involving SPE) works by collecting and then diluting the fraction(s), passing the fraction(s) through an SPE and ultimately eluting the concentrated fraction in a small amount of 100% organic solvent. An Automated Neoteric Approach to Trapping Metabolites for NMR Analysis SPE cartridges are used to trap the metabolites from natural occurring material during a repetitive analytical run with injection of the same sample; in order to concentrate the individual metabolites to a level required for NMR analysis. A Chiral Piece of the Pie-Optimizing Chiral Separations through Selective Sample Slicing and Column Switching Samples are injected onto a primary column and individual peaks are cleaved and transferred onto a secondary chiral column for separation. We demonstrate complete purification of analytes with high resolution of the separated enantiomers Increasing Throughput of GPC Clean-Up via a Parallel Sample Extraction Process The purpose of this study was to evaluate the effectiveness of developing an automated GPC clean-up system that is capable of running up to four sample extracts in parallel, along with fraction collection, thus improving sample throughput. Optimization of Steady State Recycle SSR: Parameters Utilizing Polarimetry in Chiral Separations Processing gram to kilogram quantities of target analytes has led to the exploration of several high throughput separation techniques. Among those investigated is Steady State Recycle SSR. Similar to Simulated Moving Bed SMB fractions are collected from the leading and trailing edges of a chromatographic contour while sample material is injected into the interior. The difference SMB is a continuous process while SSR is a repetitive process. Flash: Eliminating the Concentration/Dry-down Required for a Purified Compound by Preparative Chromatography Employing an activated media packed in an SPE/Flash cartridge (60 mL/10 gm), the automated system can intelligently identify the purified fractions and compound mass 0.1 2 g, without time-consuming dry-down. Fast PCLC: A widely Applicable Technique for Laboratory Scale Preparative Separations It is often expected that prep HPLC can be satisfactorily performed via low efficiency, large diameter columns, operated under overload conditions. Evaluation of this preparative HPLC, low-tech route is not the best option. Short, fine particle columns permit baseline resolution of closely eluting compounds. Incorporating gradient elution and multicycle automation, the 1-inch bore can isolate 1 mg of each of several substances, but can also be employed to obtain up to 200 grams of a single compound. This technique, called FAST PCLC (preparative column liquid chromatography), reduces analysis time, so throughput is greater. For example, a 3 um column can handle a throughput of 1 gram in about one-sixth of the time required on a 30 um column. Multiple Column Injection and Single Concentration Column Fraction collection to increase sample volume and Speed up Dry Down Presented is an application to separate flavonoids via a multiple column injection. The sample injection is optimized for peak resolution on a Prep C18 Column. Identical columns are configured in parallel for injections sequenced by the sample peak elution of the test column. Sample is concentrated on separate C18 prep columns and eluted in high organic for effective dry down of the target sample. This technique provides high throughput of sample for each cycle decreasing separation time to purify gram quantities of target compounds. FTLC: Automated Fraction trapping of Purified Compounds from Preparative Chromatography via a Specialized Polymer Resin packed within a HPLC column This trapping method allows for fractions collected to undergo transformation into 100% organic solvent without operator intervention, drastically reducing the dry down time and eliminating hydrolysis of the compound. The HPLC trapping column has been shown to be very robust and rugged stable for 1000s of fraction trapping samples.
