CHAPTER 3 - MOVEMENT OF SUBSTANCES ACROSS THE PLASMA MEMBRANE Topic :Determining the concentration of an external solution which is isotonic

to the cell sap of a plant Objective /Aim: Determining the concentration of sucrose solution which is isotonic to the cell sap of potato cylinder Problem Statement: What is the concentration of the sucrose solution that will maintain the mass of potato cylinder? Hypothesis: As the sucrose solution reach certain concentration (where it is isotonic to cell sap) ,there is no change in the mass of potato cylinder. Variables : manipulated responding controlled Apparatus : Cork borer ,test tube/specimen tube, stop watch, ruler, knife Materials : Potatoes, various concentration of sucrose solution, filter paper Technique : Measure and record the final mass of potato cylinder by using electronic balance Procedure : 1. Seven test tubes are labeled P,Q,R,S,T ,U and V. 2. Test tube P is filled with 25 ml distilled water, test tube Q is filled with 25ml sucrose solution 0.1 M, test tube R is filled with 25ml sucrose solution 0.2M, test tube S is filled with 25 ml sucrose solution 0.3M, test tube T is filled with 25 ml sucrose solution 0.4M , test tube U is filled with 25 ml sucrose solution 0.5M and test tube V is filled with 25ml sucrose solution 0.6M 3. The cork borer is pushed into the potato and the potato cylinder is obtained by pushing it out of the cork borer using glass rod . 4. The potato cylinders are cut to the exact length of 5.0 cm. 5. Record the initial mass of potato cylinders by using electronic balance. 6. One potato cylinder is placed in each labeled test tube for 30 minutes. 7. After 30 minutes, the potato cylinders are removed from the test tube and gently wiped with filter paper . 8. The final mass of the potato cylinders are measured. 9. The final mass of the potato cylinders are recorded in a result table. : the concentration of the sucrose solution : change in mass of potato cylinder : type of potato // volume of solution //immersing time
