Affinity Separation s
The technique of affinity separation is one
which has been routinely used in research and
development laboratories for the purification of a
wide range of biological materials of therapeutic
interest. In theory, affinity separation offers the
unique opportunity to "custom tailor" a separation
medium to exhibit highly specific binding to the
biological component to be purified When
reduced to practice, however, all currently available
affinity separation techniques reveal characteristics
which render them unattractive for the purification
of biological materials on a commercial scale.

Sepracor, through a unique fusion of membrane fabrication and bioengineering expertise,
has developed new affinity separation technology
which is just as amenable to commercial scale
purification of biomolecules as it is to purification
on the R & D scale. Based upon a non-fouling

hollow-fiber microporous membrane, Sepracor


technology is able to overcome the limitations in
scale-up exhibited by current affinity separation
systems .
The key to Sepracor's affinity separation
system is the hollow-fiber membrane module
shown in the accompanying photograph . This
module houses a special microporous, biocompatible polymer spun into hollow fibers of
carefully controlled dimensions (Figure 1) .
The microporous nature of the membrane has the
advantages of high surface area and therefore high
ligand loading capacity (typically 0 .5 - 1 .Og of
ligand for the device shown in the accompanying
photograph) as well as high throughput (on the
order of I liter/minute), important features when
designing an affinity separation system on a
commercial scale.

~ In fact, in comparison to the two most


widely used media for affinity separations,
Sepracor's hollow-fiber membrane device
exhibits the throughput capacity of a porous
silica column more than 100 times its size and of
an agarose gel column array greater than 750
times its size (Figure 2)! This high throughput
capacity in combination with high ligand

"loading, low pressure operation (typically 2 psi


transmembrane pressure) and the ability to easily
scale-up simply by increasing the number of
membrane modules within the system make
Sepracor affinity separation technology ideal for
the purification of biomolecules on an industrial
scale .
