La poliacrilamida posee ciertas caractersticas que la hacen til para ser usada en electroforesis: i) termo-estabilidad, ii) transparencia, iii) resistencia, iv) qumicamente inerte, v) se pueden preparar varios tamaos de poro, vi) resiste grandes gradientes de voltaje, vii) permite realizar varias tinciones de protenas, viii) se pueden extraer las protenas, ix) se puede secar

Acrilamida. Autopolimeriza espontnea pero muy lentamente. En presencia de radicales libres, el proceso se ve acelerado. stos provienen del APS en presencia de TEMED. Los polmeros que se forman, sin embargo, slo forman una solucin viscosa porque las cadenas se deslizan una sobre otra. La bisacrilamida es capaz de unir cadenas de acrilamida entre s (crosslinker), llevando a la formacin de un gel.

El SDS (detergente inico) permite independizarse de la forma y la carga de las protenas. Al quedar desnaturalizadas, las protenas adquieren cargas negativas de una manera proporcional a su peso molecular.

Ambos son agentes reductores y se usan para i) terminar de desnaturalizar protenas con puentes disulfuro intracatenarios y ii) desarmar estructuras cuaternarias mantenidas por las mismas uniones
La muestra se calienta a 95-100C por 3-5 min y est lista para ser sembrada en el gel

En el stacking, los iones Cl- marcan el frente de corrida por su alta relacin q/m. El pH del stacking -6,8- desplaza levemente el
equilibrio de la glicina (pI6,1) hacia la forma cargada negativamente, haciendo que sta migre lentamente. De este modo se genera una zona de alta resistencia, localizada entre los iones Cl- y la glicina. Dado que las protenas no pueden migrar ms rpido que los iones Cl-, sumado a que se utiliza un porcentaje bajo de acrilamida (3-5%), las protenas tienden a apilarse en esta zona de baja conductancia.

Una vez que llegan al gel resolvedor (pH 8,8), la glicina gana cargas negativas y el voltaje se hace homogneo a lo largo del gel. Desde aqu, el factor clave en la migracin de las protenas es el % de acrilamida y las protenas migran de acuerdo a su peso molecular

Detection kits For HRP-conjugated antibodies: ECL and ECL+ (home made or commercially available) are the traditional kits used and we recommend ECL+. For the new generation detection machines such as Genegnome, use the detection kit recommended by the manufacturer of the machine. We do not recommend ECL or BCIP/NBT detection kits as they are not as sensitive. X-ray films Manual film development is traditionally used and enables the scientist to control the incubation time of the x-ray film in the developing agent and fixation agent. Automated x-ray film developers are also widely used and easy to use. Remember that an over-exposed film is not suitable for analysis as determination of the relative amount of protein is not possible. Overexposed films show totally black bands with no contrast, and/or numerous non-specific bands. Digital images: The new generation of film developers are units with a camera inside an enclosure, removing the need for a darkroom. The camera detects the chemiluminescence emanating from the membrane, transforming the signal into a digital image for rapid analysis with software provided with the detection machine. A range of machines are now commercially available. At the front of the next generation are systems which do not use HRP-conjugated antibodies (i.e chemiluminescence): for example, STORM Analysers detect fluorescence from fluorochrome-conjugated secondary antibodies. The Odyssey Infrared Imaging System detects infrared fluorescence.

The LI-COR Odyssey Infrared Imaging System is useful for detecting infrared dyes. It is a two channel laser-based infrared detection system made by LI-COR Biosciences. It provides high sensitivity, multiplexing capabilities and accurate quantitation. The instrument can be used to identify fluorescent molecules that excite at 680 nm and emit at 700 nm in one channel, and molecules that excite at 780 nm and emit at 800 nm in the second channel.

The Odyssey Fc System can acquire images in both fluorescent and chemiluminescent modes. IR Fluorescence: Lasers are used to excite 700 nm and 800 nm IRDye fluorophores for 2 channel detection. Fluorescent emission is then detected to generate an image of the sample. At these IR wavelengths, membrane autofluorescence is very low. This method directly detects the fluorescently labeled conjugates. Because the fluorescent signal is very stable, samples can be archived and re-imaged. Chemiluminescence: Luminol substrate is applied to the sample, and is oxidized by horseradish peroxidase (HRP) enzyme in a light-generating reaction. This method is indirect, because it relies on the enzymatic reaction to report the location of the HRP conjugate. Chemiluminescent signals are dynamic and time-dependent. Light emission is initially strong, but diminishes over time.

Mouse liver extract (50 g) total protein was separated on a Bio-Rad Criterion gel, transferred to PVDF membrane and blocked with 1% milk in TBST. The membrane was probed with mouse monoclonal anti-Cdk1 (LabVision) (0.2 g/ml) and goat anti-mouse HRP (0.16 g/ml) or Clean-Blot IP Detection Reagent (HRP) (0.2 g/ml).

Far-Western blotting is the method of probing a membrane containing transferred protein with another protein to detect specific protein-protein interactions.
ProFound Far-Western Biotinylated-Protein:Protein Interaction Kit This kit utilizes biotin modification of a purified "bait" protein probe. Prey proteins separated in-gel or transferred to a membrane can be probed with this biotinylated bait. Detection of an interaction with "prey" protein(s) is achieved with a streptavidin-horseradish peroxidase conjugate and a chemiluminescent substrate.
