Investigating the effect of varying concentrations of biological detergent on the permeability of beetroot membranes 1.

1 Outline your biological knowledge or research related to the problem under investigation 1.2 Use appropriate biological knowledge and understanding 1.3 elect ideas from the information relevant to the investigation 1.! "ink your ideas together to suggest an e#planation The cell membrane is made up of a phospholipid bilayer which is described as a fluid, mosaic model due to the fluidity of the layer which is embedded with proteins.

The phospholipids contain a polar, phosphate head and a non-polar tail which is made up of fatty acids. The amphipathic nature of the phospholipids causes a bilayer to form

There are many proteins in the bilayer such as integral proteins, which are embedded inside the bilayer, and peripheral proteins which are attached to the outside of the bilayer. 1.$ tate precisely in a hypothesis the most appropriate e#planation I predict that an increase in concentration of biological detergent will cause an increase in enzyme activity and thus a decrease in cell membrane permeability. 2.1 Using suitable e%uipment and materials Biological detergent Used to disrupt the membrane of the beetroot cores Beetroot The biological material used in the e periment to investigate membrane permeability

!or" borer To obtain beetroot cores #azor blade To cut beetroot to a specific size $orceps To handle beetroot cores Test tubes %with labels& To contain the solution with red dye and water to use in the colorimeter !olorimeter To measure percentage light transmission 'aper towel To dry the beetroot cores Tile To place beetroot cores onto #uler To measure the length of beetroot cores (eighing scales To weigh out the mass of biological detergent used Thermometer Used to stir the biological detergent and to ma"e sure temperature is constant !loc" Used to measure the time that the beetroot cores are immersed in biological detergent and the time the beetroot cores are left in water Bea"er To

2.2 elect a suitable range for the independent variable or suggest conditions for the e#perimental treatment $or my e periment I have decided to use biological detergent as my independent variable with a range between )-*)+ by using )+ %water&, ,+, -+, .*+, ./+ and *)+ biological detergent. 2.3 &ommunicate an ordered se%uence to follow as a procedure .. (ear goggles for safety *. 'repare si detergent solution in the range between )-*)+ by following the method below0 1easure ,g, -g, .*g, ./g, and *)g of the detergent and put each substance in a separate bea"er. 2eave one bea"er with no detergent as a contro l tube Using a graduated pipette and pipette filler, measure .))ml of tap water and add to each bea"er Use a stirring rod to stir the solution to dissolve the biological detergent

3. Use a cor" borer to obtain 3) cores of beetroot ,. Use a ruler to measure ..4cm and use a razor to cut the cores to this length

4. (ash and dry the cores to remove any pigment released in the cutting process /. 'ut 4 cores of beetroot into each bea"er for .4 minutes 5. Using forceps, remove the cores from the bea"er and rinse and dry them to remove any pigment released and to remove biological detergent remaining on the cores -. 'ut each core in separate test tubes and add 4cm3 of water to each test tube. 2eave cores in water for 3) minutes. 6. 'our each solution into another test tube, ma"ing sure to not transfer the core .). 7ha"e each test tube to evenly distribute the dye ... 1easure the percentage light transmission of the solution using a colorimeter with a blue filter of wave length ,5)nm. 2.! 'rovide a fair test To provide a fair test we must ensure that certain variables are "ept constant, to ensure accurate results. In my test I have decided to "eep the following variables constant0 Temperature 8s an increase in temperature will cause the enzymes to increase in "inetic energy and thus move faster and collide more with the proteins in the cell membrane, leading to an increase in rate of reaction and more red pigment will be released Brand of biological detergent used 8s different detergents have a different number of enzymes and an increase in enzyme numbers will cause an increase in enzyme activity and thus more cell membrane bro"en down and red pigment released Time the beetroot cores are immersed in the biological detergent 8s an increase in time will allow the reaction to occur longer and so more cell membrane will be bro"en down and more red pigment will be released Time the beetroot cores are left in water 8s an increase in time will allow more red pigment to be released into the water The size and shape of beetroot cores 8s an increase in surface area will allow more proteins to collide with the enzyme and an increase in size means there will be more dye that can be released The type of filter used in the colorimeter 8s filters affect how much light and what type of light can be transmitted (ash and dry the cores to the same degree 8s dye could be left on the cores from the cutting process or biological detergent could remain on the cores after being immersed in the detergent and so the reaction would still occur thus causing more dye to be released

Use the same volume of biological detergent 8s an increase in volume will lead to an increase in numbers of enzymes and thus enzyme activity will increase leading to the cell membranes being bro"en down faster and more dye released

2.$ 'redict the result of the test of the hypothesis

3.1 &learly state what must be recorded In my e periment I will be recording the percentage light transmission of each solution using a colorimeter with a blue filter of wave length ,5)nm, and will find an average percentage light transmission for each concentration of biological detergent used. 3.2 pecify the format to be used to present the results

3.3 (ecide which method of analysis will be the most appropriate and )ustify your selection

3.! (ecide how much replication is re%uired for successful analysis $or my e periment I replicated each solution five times as seen in my method to obtain 4 results for each solution. 3.$ &onsider the feasibility of replication* considering the time and materials available The replications are very feasible as they can be created at once, as seen in my procedure above, where 4 replicates or more can be left in a bea"er of the biological detergent at once.
