2) Oxidase is not a test used for Mycobacteria, Bacillus sp. or anaerobic bacteria. I have been a
micro tech for 35 years. This is the way I utilize oxidase: If I have a nonlactose fermenter on MacConkey agar, I subculture to blood agar. I perform an oxidase test. Oxidase cannot be performed on colonies growing on MacConkey because of the color of the medium. It has to be done from a non-selective agar like blood or chocolate. If it is oxidase positive, that throws it immediately into the non-glucose-fermenter category. This is a big category and includes Pseudomonas sp. and Alcaligenes sp., as well as many other nonfermenters such as Vibrio. One must take into consideration where the specimen is from (respiratory, fecal, wound, etc.) and proceed accordingly using the appropriate key. Nowadays we use automated instruments such as the Vitek2 to run identifications on these. If it doesn't ID, we then go to other methods, such as API's NFT (nonfermenter) identification strip. There are other commercial systems available - I have just named the two that our lab uses. If I have a gram negative diplococcus, I perform an oxidase test. If it is positive, then pathogenic Neisseria sp. (N. gonorrhoeae, N. meningitidis) must be ruled out. Again, it depends on what site the culture is from. There are commercial identification systems out there for these organisms. I subculture the organism to chocolate agar and Thayer Martin agar (chocolate with antibiotics). If the organism grows on TM, then it is a possible pathogenic Neisseria and I go ahead with an identification. The only lactose fermenter I can think of that is oxidase positive is Aeromonas sp. When performing an abbreviated ID of E. coli (lactose positive, indole pos, oxidase neg) we always perform oxidase to rule out Aeromonas. BTW, Acinetobacter is NOT oxidase positive. I think it's really difficult to memorize a list of organisms by one reaction. I think it's easier to memorize the reactions that fit one organism: E. coli is lactose fermenter, oxidase neg, indole positive, motile. Kleb. pneumo is lactose fermenter, oxidase neg, indole negative, NONmotile. Kleb. oxytoca is lactose fermenter, oxidase neg, indole POSITIVE, nonmotile E. cloacae is lactose fermenter, oxidase neg, indole negative, motile, Ps. aeruginosa is nonlactose fermenter, oxidase POSITIVE, motile etc.
