Dear Colleagues, If you are familiar with the drug development cycle, and how long it takes to bring anything to the market, you would be amazed to know the progress Reliance Life Sciences - the new millennium initiative of RIL - has quietly made in a short span of less than a decade. It already has several life-saving products that India can be proud of. Traditional businesses are rarely, if at all, familiar with the enormous challenges a research-dominated enterprise such as this faces. There are special needs that academia does not provide for. There are ethical issues related to clinical research. The list is long. Read this issues lead story to learn a whole lot of things about RLS. The story may appear to be a bit long but anything smaller would hardly do justice to the subject. The MindTree story by Abhilash is highly relevant to the young and old alike in the current environment. Winds of change are blowing all around us and none of us can hide behind a fig leaf for inaction. Read and enjoy. The NewsWheel section is raining awards and Hazira tops the charts with most of these. Ananya, Aditi and Rajendra have notched up impressive individual honours too. The Editors pick is the one on Aditi and her twin brother. The Plant of the Month section has an amazing array of plants. Enjoy the issue and do let us know if you like or dislike anything. We promise to quickly respond. Happy reading!

Time 2.30 am. Date: August 24, 2013. In Gondia, a small town 160 km from Nagpur in Maharashtra, 36-year old Vinod Patil was experiencing severe chest pain. His wife frantically called up the physician Dr. Joshi, who came in shortly after. The doctor, after quickly checking Vinods pulse, informed that he had to be immediately admitted to a hospital. Soon Vinod was taken to the nearest nursing home where an electrocardiogram (ECG) was carried out. The results confirmed the doctors worst fear. Vinod, had a massive myocardial infarction (commonly known as heart attack), and, if the blood clot in the heart could not be removed immediately, he would not be saved. The nearest hospital where an angioplasty procedure could be carried out was a three-hour drive and, in wee hours, he had doubts that there would be any cardiologist to attend to him. But giving up was no option for Dr. Joshi. He made a quick call or two and sat by the patient, trying to comfort him. Within fifteen minutes a young boy rushed in with a kit in his hand. The doctor heaved a sigh of relief and, without wasting a moment, swung into action. Within an hour, the pain subsided and Vinods condition improved, much to the relief of the doctor and his family members. Vinods life was saved that night. Thanks to the promptness of Dr. Joshi and the Business Executive from Reliance Life Sciences (RLS) who ensured that MIRelTM, the lifesaving drug, was delivered at the doorstep in the middle of the night. MIRelTM, launched by RLS, is the worlds first biosimilar of Reteplase. MIRelTM is a thrombolytic agent used to treat heart attacks by breaking clots. Mahua Bhattacharya brings you a perspective on how RLS touches the lives of millions of patients like Vinod, everyday.

To the markets Biosimilars or biogenerics, as drugs like MIRelTM are commonly known, are developed after the patent protection of first generation of biopharmaceutical medicines have expired. These products closely match the innovators protein and are made with a different cell line followed by a different manufacturing and purification process. The advent of biopharmaceutical drugs have helped address unmet medical needs. However, only a fraction of the patient population has been able to afford these drugs due to steep prices they command. Over the last twelve years, RLS has built the foundation for an integrated, primarily biotechnologydriven initiative, ranging from basic and applied r e s e a r c h , pre-clinical studies, clinical research and manufacturing to marketing. RLS entry into the biosimilar domain

is driven by the larger opportunity in this space and by a vision of making these drugs available at affordable prices to the patient population. Currently RLS has focused on super-specialty therapeutic areas, such as oncology, neurology, cardiology, gynecology, orthopedics and gastroenterology. We have launched eight biosimilars in India and are now exporting some of them to semi-regulated markets too, shares Venkatesh S, Head Marketing, RLS. Today, RLS has the largest number of biosimilar products in the Indian market and is developing the largest number of biosimilars globally. It is vital not only to produce the right quality product but also deliver the right product at right time to the right place i.e. a patients bedside. In this context, supply chain plays a critical role. Supply chain management can be a potential differentiator between life and death of a patient, Rahul Padhye s i n c e R L S

manufactures and sells many life-saving drugs for critical diseases. Maintaining temperature of products during movement is of utmost importance. The transit conditions for many of these products range from ambient room temperature to 2C - 8C. The cold chain becomes an imperative in handling several of its products, shares Rahul Padhye, Business Head, Biosimilars, RLS. In the plant Manufacturing is a critical area in biopharmaceutical and pharmaceutical domains. Most of the intermittent manufacturing steps involve close monitoring and critical process control. All operations are carried under aseptic environmental conditions in classified and segregated clean zone areas. Hence, every s t a g e o f manufacturing activity is important with respect to

product yield, efficacy, stability, reproducibility and quality attributes pertaining to all commercial and clinical products, elaborates Balachandra Yalameli, Plant Head Operations, Biosimilars Business, RLS. RLS has built inhouse manufacturing capacities and all these facilities are compliant with United States Food and Drug Administration (USFDA) and European Medicines Agency (EMEA) standards. It is currently operating the largest mammalian cell culture facility (10,000 litres capacity) in India in Navi Mumbai. RLS has forayed into the pharmaceutical domain and the team is focused on developing active pharmaceutical ingredients (API) and oral solid dosage and injectable formulations of o n c o l o g y products for domestic and global markets. U n l i k e biosimilars, RLS faces relatively more competition from well-

Santhosh Mathai

established Indian companies in the pharmaceutical domain for both API and formulations. It is imperative for RLS to launch products in developed markets and maintain uniform quality standard globally, informs Santhosh Mathai, Business Head, Pharmaceuticals, RLS. The state-of-the-art manufacturing facilities, compliant with USFDA and EMEA guidelines, are equipped to deliver high-quality products consistently and at competitive prices. The challenge faced i n t h e pharmaceutical process include the use of c o m p l e x c h e m i c a l technologies, h a z a r d o u s Jayant Mundle chemicals, containment and environmental issues, says Jayant Mundle, Head API Manufacturing, Pharmaceutical Group, RLS. The process yields and quality of products are governed by precision of process parameters and environment conditions in the

manufacturing f a c i l i t y. W e ensure that the production and utility equipment are always i n b e s t condition and environmental requirement are met. This poses a Sandip Mane challenge since tolerances are too low, says Sandip Mane, Head Engineering Group, RLS. Plasma Proteins is another category of life saving drugs. Plasma proteins have human origin and there is no known substitute for human immunoglobulins which makes these products indispensable in critical care situations. RLS is the only plasma proteins manufacturer in South Asia. Since RLS commands leadership position in plasma proteins market, quality is of paramount importance for long term sustainability. RLS continually upgrades its manufacturing facilities to meet the current Good Manufacturing

Process (cGMP) standards. In plasma proteins group, the training manager o r g a n i z e s continuous l e a r n i n g programs for operational staff to keep them Vinay Ranade abreast of current cGMP practices. All products are manufactured and tested as per stringent specifications of European Pharmacopeia, informs Vinay Ranade, B u s i n e s s Head, Plasma Proteins, RLS. P l a s m a production is a complicated process. By controlling pHv a l u e a n d temperature, proteins can be precipitated one

after the other. There may be slight deviation in measuring the pH of the solution by normal pH meters, so we have to draw a sample, adjust the pH and accordingly replicate it in the major batch, explains Dr. Rainer Pabst, Head Manufacturing, Plasma Protein Group, RLS. For plasma p r o t e i n manufacture, sourcing raw material is a challenge, since the source is of human origin. We have tied up with several Indian sources to Dr. Manjunath Kamath ensure a steady supply of raw material, shares Dr. Manjunath Kamath, Head Production, Plasma Protein Group, RLS. In plasma protein manufacturing utmost emphasis is given to both product and operator safety and rigorous gowning procedures are followed for any person entering the production area, says Tabita Jairaj working as a Manager in Plasma Proteins Manufacturing Group at RLS. Qualitycornerstone

Quality in life sciences domain is critical as products manufactured in the plants impact human lives directly. Quality begins at R&D stage, hence all developmental activities should have robust method and processes in the place right at the beginning, which is done by a d o p t i n g systematic, proactive and Veena Shetty s t r a t e g i c

methodologies, shares Veena Shetty, Head Analytical Development Formulations R&D, Pharmaceutical Group, RLS. The products developed appropriately at the initial stage would have lesser problems during technology transfer, thus saving time and money for the organization. Even in the manufacturing phase, we have to be extremely quality-conscious as the safety and efficacy aspects of products c a n n o t b e compromised at all, says Devu Raj working as Manager in fillf i n i s h Devu Raj manufacturing facility at RLS. The major challenges faced in variability of results are due to the biological nature of the products. Endotoxin testing is a challenge as we try to meet stringent limits and comply with international regulatory requirements. The major focus of our team is to meet testing requirements of various regulatory bodies

across the globe, as RLS expands i t s g l o b a l footprint, shares Dr. Jyoti Patel, Head Quality Control Group, RLS. RLS has adopted a policy of non-tolerance towards any nonDr. Jyoti Patel compliance to SOPs to bring about a cultural shift, which is critical in establishing 24 X 7 audit readiness in the organization. Clinical Trials In any drug development process clinical trials are an integral phase. R L S h a s instituted a clinical research organization to meet clinical

research requirements of RLS products as well as provide full-range of services to meet study needs of other pharmaceutical and biotechnology companies. This has enabled better control on quality and time and also growing the knowledge base, says Jamila Joseph, Head Clinical Research Services Group, RLS. At the bio-analytical laboratory, the team develops methods for analysis of sample as well as drugs used in clinical trials designed to evaluate safety of drug products for human consumption. These trials/studies are called pharmacokinetic studies where we measure accurately the amount of drug of interest and/or its associated metabolite/s from study samples after the drug (under trial) is administered to healthy human subjects/patients, explains Aslam Khan, Head, Bioa n a l y t i c a l Laboratory, RLS. Before the drugs are tested on volunteers it is Aslam Khan important to test

the safety of these drugs in similar systems m i m i c k i n g humans, in this case it is animals. For example if we are working on an oncology drug, the human cancer cell is Dr. Harinarayan Rao introduced in an immuno-deficient animal, it co-lives with the cell lines, develops into a tumor which can be treated by an oncology drug, this helps us to understand the dynamics of drug mechanism. Animals are bred for product specific need and same two models never work for a product, this is both an excitement and a challenge in this job, explains Dr. Harinarayan Rao, Head, Preclinical Services, RLS. In the R&D lab The research and development function forms the genesis of any life sciences organization. Any organization operating in biotechnology space can sustain only if innovation is its major

growth driver, s h a r e s D r. V e n k a t a Ramana, Chief Scientific Officer, RLS. RLS is developing novel molecules to address unmet medical needs in areas of dengue Dr. Venkata Ramana f e v e r, c a n c e r treatment, infectious diseases, inflammatory disorders, ocular disorders and neurodegenerative disorders. The discovery program is based on small interfering ribonucleic acids (siRNAs), monoclonal antibodies and fusion proteins. Further, RLS has in-house capabilities for endto-end developmental activities, starting from cloning of the gene, expressing the protein and finally zeroing in on the process giving the best yield. The key challenge in biologics process development is that biosimilars are far more complex structurally, hence, more difficult to characterize, produce, and reproduce. The process parameters for biosimilars are defined early with a limited scope for later revisions.

Further, organizations like RLS have taken a step forward to entering the era of predictive and preventive diagnostics wherein disease

conditions are identified much before they c l i n i c a l l y manifest. We are now moving to an e r a w h e r e diagnostics would lead us to understanding a patient's genetic Rajesh Korde makeup and guide therapeutic selection. Specific abnormalities like cancers, infectious diseases, genetic and metabolic disorders are tested through DNA, RNA and protein identification and quantification, elaborates Dr. Rajesh Korde, Head Molecular Medicine Group, RLS. Targeted therapy which helps to find the right fit for the right patient, is another area in which RLS is working on. RLS has been a pioneer in India in the area of preimplantation genetic diagnosis (PGD), whereby the team can do genetic testing on in-vitro generated embryo and identify the disease-free embryos, for transfer to mothers womb.We offer prenatal diagnosis, a highly challenging job, to decide whether the unborn child is free

of the genetic disease under investigation. This testing needs to be carried out by technically skilled personnel as we handle fetal samples. before boarding the train. I ignored this guy and continued gazing out of the window, looking at nothing but the dark open fields. And then, I did my first blunder. I mistakenly looked into his eyes. You want some? he asked, giving me no time to escape. Ah, no, no. Thanks, I said after realizing that he was actually offering me some chips and wafers to eat. If youre hungry, you can have it. No need to hesitate, his voice was shivering and smelt awful. I nodded in a no again. He stared me as if he needed an explanation. I gave a fake smile and said: Ah, actually, I dont take anything to eat or drink from strangers.

IIT! Good, good! I too have some niece in some IITI dont remember which though. After seeing his high state, I certainly didnt expect him either. You guys can do well. Certainly! he added. Yah. Yah! I again tried to chicken out. No, you see, these frauds who have covered themselves with the shield of government tag have ruined this state. You must have read history. Bihar and Bengal were once the land of treasures in this country. For years and years, vultures came in and tore away every bit of beauty from here. All that is left is backwardness, poverty and illiteracy. Ah, come on, I thought to myself, as I was the only listener of this drunken mans lecture. He continued: Have you ever been to villages in the interiors of Bihar? You may have seen people there. The only concern in their life is hunger. They never even expect to have education or electricity or anything like progress. Their only saviours are young people like you who have the tools of knowledge. You must know the silly progress plans of this new government, dont you? In reality, I knew nothing. I nodded in a no and hesitatingly said: Ah, actually I am not interested much in these affairs. I have my own areas of interest And he sat silent for about ten seconds, dumbstruck, staring at me. I felt embarrassed. He chuckled with himself and asked: Did you ever in

your life thought about entering politics, son? No, sir, I am not that imprudent and I have cleaner options in life. He sighed and turned his head towards the window in despair. His expressions reflected disappointment. Then, I asked: What do you do sir? Oh me, Well, I have been in politics since I was 22. We built a new party which aimed to involve the youth of this nation. Thats nice! But you were in politics, means not anymore? What happened? Yah, the party eventually shattered. Today I am

unemployed and seek for some work here and there. Party shattered? How? He went silent. He didnt utter any word for a few seconds. He was submerged in some thinking. After the pause, he said: Son, in this country of billions, we still have scarcity of young legs to walk the talk when it comes to politics. Its true; you can change the nation but not its people! My words were gone. He turned towards the window and gazed outside. Then he loudly burped, a burp of disgust, and never spoke to again.

surface. Even the drought tolerant annuals will bloom better if they get regular water. Soil A few annual flowers prefer rich loamy soils and others do best in sandy well-drained soil. Before you dig deep into the ground to plant seeds / seedlings, remove weeds. Mix wellrotted cow dung or vermi-compost and farm yard manure at the time of preparation. Add some amount of insecticide along with a small quantity of neem cake for healthy growth. Care and upkeep Though most of the annual plants require low maintenance, you must know about the basic necessities and special needs of your plant. Many annuals require dead-heading, removing flowers once they begin to fade. A weekly walk through the garden deadheading spent blooms will keep many annuals flowering longer and pinching. Removal of apical shoot is essential to promote growth of laterals and suppress apical dominance. Maximum growth occurs in the months of September to March. Applying a balanced NPK fertilizer once a month will help the plant luxuriantly and quickly grow. NPK fertilizer is primarily composed of

three main elements: Nitrogen (N), Phosphorus (P), and Potassium (K), each of these is essential in plant nutrition. Do not forget to prune, weed and remove the spent flowers to retain healthy looking coloured flowers to welcome and inspire you every time you visit your garden next! Happy Gardening!
