PRELIMINARY ACTIVITY

FOR PRIMARY PRODUCTIVITY


Purpose:
- To test the organisms whether the light effect to their photosynthesis or not.
- To check rate of organisms of use and produce oxygen for respiration and photosynthesis.

Hypothesis:
The more light has, the more oxygen there are in the water because if you have more light, you have
more photosynthesis and that photosynthesis produce oxygen but if you do not have light, you do not have
photosynthesis then the oxygen in water will decrease because of respiration of organisms.

Materials:
- Water with organisms
- Foil
- Light
- Dissolved Oxygen Probe

Procedure:
1. Prepare the Dissolved Oxygen Probe for use following instructions from your teacher. Place the probe in
a beaker filled with about 100 mL of distilled water.
2. Connect the Dissolved Oxygen Probe to the interface and open the data-collection program. Allow the
probe to stay in the water for 5 minutes as the probe warms up.
3. Collect DO data.
3.1. Place the tip of the probe into the water sample being tested. Submerge the probe tip to a depth of
46 cm.
3.2. Start data collection. Gently stir the probe in the water sample. Note: It is important to keep stirring
until you have finished collecting data.
3.3. Continue stirring and data collection until the readings have been relatively stable (stable to the
nearest 0.2 mg/L) for about 30 seconds, then stop data collection.
3.4. Select the stable region of your graph, then display Statistics for that region. Note and record the
mean value for that region as the DO of the water sample.


Results:
- The bottle that has light get 5.474 mg/L dissolved oxygen.
- The bottle that does not has light get 6.001 mg/L dissolved oxygen.


Discussion:
In this experiment, we have the hypothesis that the oxygen in the bottle that have nothing cover will have
more oxygen than the one that have some material cover. But when we did the experiment, the outcome didnt
follow the hypothesis that we had predicted because there are more oxygen inside the bottle that have material
cover than the bottle that have nothing cover. We predict that this error happened because we didnt check the
organisms inside the bottle because the organisms in both bottle are may not equal or do not have enough light
for they to have photosynthesis so our outcome is error so for the next experiment we will check the organism in
the both bottle before we have experiment. In the next time we can use this result to have many experiment such
as can the organisms produce the oxygen instead of using the machine.

1. What was the DO of the water sample you tested in the Preliminary Activity?

2. Only respiration can occur in a dark bottle. Respiration rate is the decrease in DO over time. Suppose a
dark bottle sample of the water you tested above was tested 24 hours later and found to have a DO level
1.2 mg/L lower then the initial DO value you obtained. Calculate the respiration rate (in mg/L/h) using
the formula

























3. Suppose a light bottle sample of the water you tested above was placed next to a light source. Suppose it
was tested 24 hours later and found to have a DO level 0.5 mg/L higher then the initial DO value you
obtained. Calculate gross productivity (in mg/L/h) using the formula.





































4. Calculate net productivity (in mg/L/h) using the formula.
