BIO-PLEX UNIVERSITY™ COURSE DESCRIPTIONS

2-Day Introduction to Bio-Plex Training Course ($2500 per person)

Two-day course conducted at Bio-Rad in Hercules, CA or Waltham, MA. Course designed to provide
familiarity with xMAP principles and practices in general, and for Bio-Plex in particular, to those new to the
technology.

Protein and/or nucleic acid-based assays are covered, including hands-on wet lab work and subsequent
data analysis. Experimental design topics are discussed, including a review of best practices for
processing different types of sample matrices. Principles of multiplex immunoassay data analysis are
taught including curve fitting, recovery, and determination of observed concentrations.

Course includes considerable emphasis on Bio-Plex Manager software capabilities, as well as information
on various other xMAP software packages. Start-up, calibration, validation, and shutdown, as well as
routine maintenance and troubleshooting are highlighted.

Commercially-available kits are reviewed, as well as custom assay development topics for both protein
and nucleic acid work. Conventional polystyrene beads are compared and contrasted with magnetic
beads. Course will include use of vacuum manifold as well as automated bead washer.

This "Level I" course can function as prerequisite for the 3-Day Advanced Bio-Plex Training Course. The
Level I and Level II courses can be taken consecutively. Maximum six attendees for Hercules, maximum
four attendees for Waltham.

3-Day Advanced Bio-Plex Training Course ($3500 per person)

Three-day “Level II” course conducted at Bio-Rad in Hercules, CA or Waltham, MA, designed for
experienced Bio-Plex users. Experienced IS 2.3/xPONENT users may attend, pending approval by
instructor. Minimum prerequisite is the Level I course or equivalent experience.

This course carries a strong focus on user-specific assays/analytes in a hands-on, wet lab format.
Trainees are encouraged to bring their own samples to the course. Best practices for processing different
types of sample matrices are reviewed.

Course can be tailored for cytokine (protein), phosphoprotein, or nucleic acid-based assay tracks.
Introduction to custom assay development for proteins or nucleic acids on either polystyrene or magnetic
beads is an optional alternative track.

Sound experimental design is emphasized, focusing on how to optimize for maximal sensitivity and
dynamic range with minimal and reproducible intra/inter-assay CVs. Topics include establishment of
limits of detection (LOD) and lower/upper limits of quantitation (LLOQ, ULOQ) to define an assay’s
effective working range.

In-depth multiplex data analysis topics for protein tracks will include principles of relative quantitation,
advanced curve-fitting techniques/algorithms, and comparison of xMAP data to ELISA and other
immunoassay technologies. Training on Bio-Rad’s SNP Manager software is provided for the nucleic
acid track.

Maximum attendance for this intensive course is three for Hercules, two for Waltham, in order to
maximize teacher:student ratio, except by special arrangement with the instructor.
