Identification of Caffeine in Coca-Cola by using HPLC

ABSTRACT
Has conducted research on the levels of caffeine in soft drinks in cans or bottles by using
liquid chromatography method or KCLT high level [1]. most use of caffeine aims to meet
needs in the industry drinks and also used as a flavor enhancer or seasoning on some
food flavoring (Misra et al, 2008) in the present study will establish the caffeine content
using HPLC method

INTRODUCTION
Along with the increasing
growth in the food and beverage
industry. In soft drinks often added
caffeine, artificial sweeteners and
preservatives whose levels must be
considered because if left unchecked
can cause disease in the human
body[2]. Soft drinks (soft drinks) is
defined as non-alcoholic beverages,
both containing carbonate (soda) or
not containing carbonate. In general,
a soft drink made from water, sugars,
organic acids and other auxiliary
substances, and to drink carbonated
soda is also added. Especially for
carbonated soft drinks cola type
usually also added caffeine [1].
Caffeine
consumption
,
especially in the increasingly popular
among
students
because
of
the
stimulation effect of helping students to
learn hearts long period of time ,
especially when the workload increases
as before a test or exam .

The addition of caffeine in


carbonated soft drinks cola type
intended to evoke a sense of fresh,
pep, also to relieve fatigue and
sleepiness. This can occur because
caffeine has a stimulating effect on
the central nervous system and the

skeletal muscles. In addition, caffeine


also can stimulate the heart and
respiratory system [1].
in this study , using the HPLC
method . Another method used to
determine the caffeine content using
paper chromatography method , but
it is rarely used Considered therefore
are no longer modern and efficient .
so this method is rarely used for
routine analysis [2].
MATERIALS AND METHODS
Method
In this study using HPLC
method using a set of tools HPLC.
there are several ways to define the
levels of caffeine such as using
spectrophotometric
method
,
iodometric
and
paper
chromatography, but because it is
not very efficient for everyday assay.
can use HPLC method

As much as 5 nanometers
standard solution of caffeine injected
into
the
HPLC
injector.
The
composition of the mobile phase

used was methanol: water at a ratio


of 100: 0, 90:10, 80:20, 70:30, 60:40,
50:50.
Determination of Rate of Water
At this stage used the
optimum mobile phase composition.
A total of 5 nano liter caffeine
standard solution is injected into the
HPLC injector, with a variety of power
flow rate of 0.5; 0.8; 1; 1.25 and 1.5
ml / min.
Determination of pH

standard caffeine injected into the

obtained in previous studies[2].

CONCLUSION
based on the data it was
concluded that to determine the
caffeine content in the sample can
use
HPLC
method,
some
optimizations performed on HPLC
systems can improve analytical
performance, all of which showed the
effectiveness and sensitivity of the
methods used.
