laboratory supplies and equipment. The TOXI-Lab TLC plates, TOXI-Gram, are
composed of glass fiber paper impregnated with silica gel or a C-8 (carbon aliphatic)
reversed-phase sorbent. Drugs are extracted from urine, stomach content, or other
specimen in prepared TOXI-Tubes containing buffer and an optimized extraction
solvent mixture. Sample extracts are evaporated in disposable aluminum cups with small
disks the same material as the plates. As the evaporation proceeds, extracted drugs are
adsorbed into the disk, which, when dry, is inserted into a matching hole at the bottom of
the plate. Standards are contained in similar disks. Both prestandardized and
unstandardized plates are available and standard impregnated disks are available
separately.
The prepared plate is developed in a glass chamber with a solvent mixture
designated by the manufacturer for each chromatography system. After developing, plates
are dried, and then drugs are visualized by sequentially dipped the chromatogram into a
series of tanks containing reagents and viewing under king-wave ultraviolet (UV) light.
Colors and positions of spots are recorded at each visualization stage. Photographic
illustrations of visualization stages and metabolite patterns are available in a compendium
comprising more than 100 drugs with new drugs added as data are developed.
An available PC-compatible search program can help with data analysis, yielding
statistical probability for identifications. Systems are available to screen for basic and
neutral drugs, acidic and neutral drugs, tetrahydrocannabinol metabolites, and opiates, as
well as the C-8 reversed-phase system that is used for further differentiating and
confirming presumptive findings. In addition, confirmatory procedures for many drugs
are described. The TOXI-Lab system is a powerful tool for screening urine and stomach
contents. It is less effective for blood and other tissues due to interference form lipids and
sensitivity limitations.
1.1 TOXI- LAB A
1.1.1 Introduction: TOXI- LAB A Product Description
The TOXI LAB A-PLUS System is a rapid thin-layer chromatographic method for the
detection of basic and neutral drugs. The TOXI LAB A-PLUS System is complete with
hardware and supplies for laboratories that require high volume throughput or analyst
specific analyses. Urine is added to TOXI-Tube A which contains an organic solvent and
buffers to bring to the ph. of 9. After extraction, the organic phase is concentrated onto a
TOXI-Disc. The TOXI-Disc is then inserted into a TOXI-Gram A. This is a thin glassfiber sheet impregnated with silica gel mans a vanadium salt. The TOXI-Gram also
contains a number of standard drugs, in position 1, 2, 3, 4. The chromatogram is then
developed with methanol : water : ethyl acetate (2:1:58), plus a specified volume of NH4OH.
After drying the chromatogram, the drugs are visualized by a staining technique. The
chromatogram is initially exposed to formaldehyde vapor and then dipped in concentrated
H2SO4. This is followed by a water wash and then examination under UV light. The final dip
is in a modified Dragendroffs reagent (Iodinated bismuth sub nitrate). The unknown is
compared with the various standards, at all stages of the visualization process. The results are
recorded on a TOXI-Lab A Worksheet.
Reagents and Equipment Required:
1. TOXI-Lab A Kit:
a. TOXI-Gram A. These have a V printed on top to indicate that they contain
vanadium.
b. TOXI-Disc A. For basic extract concentration.
c. TOXI-Tubes A. Extraction tubes.
d. TOXI-Dip A. Modified Dragendroffs reagent.
e. TOXI-Dip A jars.
f. TOXI-Lab A Worksheets.
2. Developing Solution A
Mix 3 ml of methanol, 1.5 ml distilled water and 87 ml Analar ethyl acetate. Shake for 20
-30 seconds.
3. TOXI-Dip A-1
Add 10 12 ml of formaldehyde solution (= 37% HCHO) to the bottom of the A-1 jar.
Blot dry the base plate and cap tightly.
4. TOXI-Dip A-2
Fill the jar to about 5 mm from the top with concentrated sulphuric acid. Place the jar in
the base of a large Petri dish.
5. Beaker or Jar filled with Distilled Water.
6. TOXI-Dip A-3
Add the contents of the A-3 vial and qi ml of glacial acetic acid to the A-3 jar and mix.
Add distilled water to bring the level to about 5 mm from the top. Cap tightly and mix
well.
1.1.2 TOXI- LAB A Procedure
NOTE: The TOXI-Grams should only be handled with the plastic forceps provided.
Preliminary Steps
i.
Plug in the electric warmer.
ii.
Into separate wells of the cool evaporation plate, place 2 A Blank Discs.
Extraction
i.
Shake a TOXI-Tube A to dislodge the salt.
ii.
To the TOXI-tube A, add urine to the pour line (5 ml arrow for TOXI-Tube A). Cap

and mix the tube by inversion for approximately 2 minutes. (Do Not Shake)
Centrifuge the tube for 2 5 minutes. (The colored aqueous layer should be on the

Development
i.
Transfer 3 ml of A developing solutions into the respective chromatography jars.
With a positive displacement sampler, transfer the volume of ammonium hydroxide,
recommended on the TOXI-Gram label, directly to the fluid in the specific
chromatography jar. Cap immediately and swirl each jar vigorously for a few

seconds.
Remove the TOXI-Gram from the warmer and lower each, disc ends first, into the
specific chromatography jar. The side edges of the TOXI-Gram should now touch the
jars. Cover and do not disturb during migration.
iii.
Remove the TOXI-Gram when the dye spots reach the level of 0.95 (13 18 minutes)
and place on the warmer face down for 20 30 seconds, until the fumes have
vanished.
iii.
Stage II
Dip the TOXI-Gram briefly in and out of a jar or beaker of distilled water. Wait about
5 seconds and then dip again. Some drugs will become visible for the first time,
others will fade, and still others will change color. Attempt to match the position and
color of any spot in the central unknown zone with a standard drug having the same
characteristics. Record your observations on the TOXI-Lab A Worksheets. Carry out

one further dip in the water and note any changes in the spot.
Stage III
Re-dip the TOXI-Gram several times in the same jar of water and then view, in the
dark, under long-wave ultraviolet light (366 nm). Use both transmitted and reflected
UV light. Attempt to match the position and fluorescence of ant spot detected in the
central unknown zone with a standard drug having the same characteristics. Record

an acidified solution (pH4.5)into an extraction solvent (methylene chloride and heptane with
zinc chloride to facilitate the extraction process). The solvent is concentrated on to a TOXIDISC Blank B. The dried disc is placed onto a TOXI-GRAM B for elution in a developing
jar. The resulting positions of drug of interest are visualized by dipping the TOXI-GRAM
into a series of solutions. The preliminary identification is based on matching the position of
a drug (Rf) and visualization color characteristics with that of correspondingreference
material.
1.2.2 TOXI-LAB B Procedure
This is a summary method for theTOXI-LAB TOXI-B thin layer chromatography (TLC)
drug detection system. The system is used to screen for the presence of a wide variety of
acidic and neutral drug compounds in urine. The TOXI-B system provides a preliminary
result that must be confirmed by GC-MSD.
Methods:

Extraction
Label TOXI-TUBES B for negative control, positive control and appropriate
laboratory numbers. Transfer 4.5mL of casework, negative and positive urine to
appropriate TOXI-TUBES B. Rock TOXI-TUBES B for a minimum of 2 minutes.

Transfer 3mL elution solvent to chromatography jar. Add volume of ammonium
hydroxide indicated on TOXI-GRAM B jar. Cap and swirl vigorously for a few
seconds. Place TOXI-GRAM B into chromatograph jar and cover. Make sure to not
allow the side edges of the GRAM to touch the walls of the jar. Allow solvent to
migrate until the dye spots reach 9.5 cm. Remove the GRAM and place face down

Dip the GRAM in and immediately out of TOXI-DIP B-1. Allow GRAM to sit until
all of the dichloromethane has evaporated. Dip GRAM in and out TOXI-DIP B-2 jar.
Note color and position of reactions for specimen spot(s). After a golden brown
background develops, place GRAM into TOXI-DIP B-3 jar and agitate until the
background clears. Note color and position of reactions for specimen spot(s). Place
GRAM into sheet protector and copy with laboratory photocopier. Lightly blot
GRAM with paper towel to remove excess reagent. Observe GRAM under UV light
(365nm). Compare fluorescence of specimen spot(s) with reference drug spots. Note

2. Concentration - Place a blank disc in a concentration cup for each sample. Transfer the
upper (organic) layer from the extraction tube into the cup. Evaporate the organic layer
to concentrate the analyte onto the disc.

1.3.1 Pendahuluan : Toxi - LAB AB Deskripsi Produk

VII. Kriteria Identifikasi
Posisi (Rf) dan karakteristik warna pada setiap negara bagian visualisasi tempat terkenal karena
spesimen harus sesuai dengan yang bahan referensi.
