Lab 1: Blood Typing

Purpose
The purpose of this lab is to perform a slide abo/rh blood typing procedure, recognize
the clinical significance of abo/rh blood typing, and how antigens and antibodies relate
to the blood typing procedure.
Hypothesis
If proper blood typing is not done, dangerous outcomes can occur. Not all blood types
are compatible.
Experiment
Materials:

Anti-A serum
Anti-B serum
Anti-RH serum
Six blood samples
Stirring sticks
Six blood typing slides
Light
Biohazard container

Procedures:
1. Place one drop of blood in each well on slide
2. Place one drop of Anti-A, Anti-B, Anti RH serums in designated wells on
slide
3. Stir each well with a separate stick and dispose of properly
4. Place under light, and determine antigens/antibodies present
5. Record in table

Your answer: With blood type AB, the appearance was A, clumpy; B, clumpy; Rh, un-clumped.
My predictions were wrong.

Your answer: sample 4 because it was O- the anti-serum with A was negative. B was negative.
RH was negative.
6. What antibodies would be found in the plasma of blood sample 1?
Your answer: Antibody A
7. When transfusing an individual with blood that is compatible but not the same type, it is
important to separate packed cells from the plasma and administer only the packed cells. Why
do you think this is done? (Hint: think about what is in plasma versus what is on RBCs.)
Your answer: the plasma contains the antibodies that could react with the recipients antigens on
RBCs.
8. List the blood samples in this activity that represent people who could donate blood to a person
with type B+ blood.
Your answer: sample 2: B+, sample 6: B-, sample 4: OConclusion
In conclusion, my hypotheses was correct, not all blood types are compatible. Also, interestingly the
most common in all ethnic groups is Type O. Least common type in all ethnic groups is Type AB.
Types that vary due to ethnic origin are Type A and Type B.
