Beers law states that the intensity of a beam of parallel monochromatic radiation
decreases exponentially with the number of absorbing molecules. In other words,
absorbance is proportional to the concentration. Lamberts law states that the intensity
of a beam of parallel monochromatic radiation decreases exponentially as it passes
through a medium of homogeneous thickness. A combination of these two laws yields
the Beer-Lambert law. Beer-Lambert law occur when beam of light is passed through
a transparent cell containing a solution of anabsorbing substance, reduction of the
intensity of light may occur.
Quantification of medicinal substance using spectrophotometer may carried out by
preparing solution in transparent solvent and measuring its absorbance at suitable
wavelength. Scan standard solution in UV spectrophotometer between 200 nm to 400
nm on spectrum mode, using diluents as a blank. The wavelength for Paracetamol
normally selected is wavelength of maximum absorption (max) at 257nm, where small
error in setting the wavelength scale has little effect on measured absorbance. Ideally,
concentration should be adjusted to give an absorbance of approximately 0.9, around
which the accuracy and precision of the measurements are optimal. The assay of
single component sample, which contains other absorbing substances, is then
calculated from the measured absorbance by using one of three principal procedures.
They are, use of standard absorptivity value.
The absorbance reading was recorded at 0.2110A. Based on the calculation on the
content of paracetamol in each tablet is 259mg consist of 59% of the claimed amount.
There are slightly different compare to the concentration of paracetamol obtained in
the experiment to the stated amount of the commercialized paracetamol which is in
500mg each tablet.
An experimental method may be accurate but not especially precise if it suffers
from random errors, but equally it may yield inaccurate results with high precision if
it is subject to systematic errors. The systematic errors affect the accuracy of the result
and will tend to do so in the same direction. The results will always tend to be too
high or too low rather than fluctuating about some central value following successive
measurements. To improve the experimental and get desired results need to free from
errors whether it systematic or random errors that might happen during experiment. It
also can be improve the precision and accuracy results considerably by increasing the
sample size. So that the experiment might have more precise and accurate results.
