Biology Lab Report #2: How Temperature Affect Enzyme Activities

Introduction:

Enzymes are the biological catalyst, which means they can increase the speed of a
biological reaction and make the reaction faster.
_Quoting from reference.com: "Enzymes are special proteins that control the speed of

certain functions in the body; without them, chemical reactions would take place too slowly
to keep a person alive. There are many different types of enzymes in the human body."
Examples: substrate(s) -enzyme- product(s)

Factors that affect enzyme activities are temperature, Ph,

The purpose of this lab experiment is to show how temperature affects enzyme activities.
The rate of an enzyme-catalyzed reaction increases as the temperature is raised.

Materials:
_Raw liver
_Petri dish
_Dropper Pipette
_1% h2o2 solution
_liver puree
_25ml graduated cylinder
_5 50ml breakers
_filter-paper disks
_forceps
_glass-marking pencils

_ice bath
_ 3 thermometers
_warm water bath
_clock with second hand
_paper towel
Room temperature-control
Hot/cold-variable

Procedure:

Use lab safety equipments (gloves, goggle).

Use scissor to cut the all the filter paper into smaller sizes.

Take the thermometer and put it the liver and wait the red thing in the thermometer stop
then write down the temperature for the liver.

Take the filter paper and dip it into the liver but only a little, not too much.

Take the filter paper that has been dip back to your table.

Take the forceps and then put the filter paper into the hydrogen peroxide using the
forceps

Take out the clock or your phone to take timer

Write down the time for that temperature that you just did

Do from step 3 and down again but now for another paper.

Data & Result:

+Cold water: It has more bubbles but it is floating on the water slower than others.
+ Hot water: It floats on the water faster than others but has less bubbles.
+ Room temperature: It is the one that has the most bubbles and it floats kind of fast. Its
reaction is the strongest.

Discussion:

As you can see our graph the hot one is the fastest, the room temperature is the lowest,
and the cold one is the middle. We need to graph datas to help everyone see it more
easier to study and research about it. The graph helps viewers easier to visualize and
understand datas. It will might be helpful for whom wants to do the experiment like us.

No, it doesnt support my hypothesis because at first I think the trend will go up but it go
up a half and go down.

If it could be an error, the error is when we calculated the time wrong. Example like we
stop the watch slower than the reaction 2 seconds. It will make our datas wrong.

We have to repeat it three times because we need to take the average of 3 of them. It will
help us know was we wrong or right.

We need to take the average because it will make the result more accurate. Example if the
temperatures are different from each time you could no your experiment has error
somewhere and you could fix it. After all the significant reason is about the accuration.

Research question:

Enzymes speed up chemical reactions that take place in cells. To do this they provide
sites where reactants can be brought together to react. These sites reduce the energy
needed for reaction. Enzyme substrates make it even easier for reactants to be brought
together and reduce the activation energy. (Referred to as the lock and key because the
substrate binds to the enzyme site fit together so perfectly) (Quizlet, 2014)

A change in pH could change the shape of hexokinase. This change would diminish or
possibly eliminate the ability of glucose and ATP to bind to the active site on the enzyme.

Reflection:
I've learned two important skills from this lab these are: calculation and how to work
smoothy with partners in lab. Such as while Tom took the raw liver I will prepare for the
hydrogen peroxide.
I hate the smell from this experiment. It make my noise nearly uncomfortable. But I was
enjoying while we watched the reaction. The test was doable.
In my opinion, I think my group work organized because Tom and I were timing exactly
the time we brought the raw liver and the hydrogen peroxide back to our station so that we could
do the experiment faster.
We need to improve our lab skill and our members should focus more while we do the
experiment
