Substrate Concentration
and pH Affect EnzymeControlled Reactions

Introduction:
According to Mhhe, Lactase is enzymes that break down the sugar lactose and make it easier
for our body to absorb the chemical such as glucose and galactose. There are some people that
have a special case of lactase which are known as lactose intolerance. These people dont have
enough lactase in their body. An individual can eat a certain amount of lactose, if then consume
too much, it will not has enough lactase enzyme to break down the lactase. If the lactase doesnt
get breakdown properly, it will creates sickness such as cramps, bloating, gas, and diarrhea.

Discussion:
As so can see from the table and graph. Yes this is a linear relationship. If you increase the
substrate concentration increase the initial reaction rate of the enzyme-catalyzed reaction also get
increase. With the more amount of substrate that you put in the more reaction will occur because
there is more substrate to be use up. When the initial reaction rate increase the process also goes
faster.

The maximum initial reaction rate for the lactase enzyme at pH 7 is 350. You can see the result
when you put 4.0g of substrate and 8.0g of substrate concentration into pH 7. You can also see
the result on the data table and graph.

The maximum initial reaction rate cannot be reached at low lactose concentrations because there
will not be enough lactose for the enzyme to react. This is also proving for the relationship
between the substrate concentration and the initial reaction rate of the enzyme- catalyzed.

The data that I get from this lab experience can indicate about the pH level for the lactase
catalyzed reaction. From my data, I can see the optimum pH level is 7.

Temperature is factor that can affect the enzyme reaction. If the temperature is too hot, the
enzyme will get kill or unable to function normally. If the temperature is too cold the enzyme
will react very slowly. This suggest the importance role of the temperature, the enzyme will
work more efficiently in the right temperature and environment.
In my opinion, the fact that the pill form of the enzyme would have to travel through the persons
stomach required many special consideration that the producer of this product need to be
concerned about is that the amount of the acidic or base. They also have to considered about the
way that the person consumed the pill, they make not take it in properly or cannot digest it right
way. The lactase of pH in the pill should be neutral because, the person may not get the enzyme
amount that they want.
