A. Title
Induction ovulation of Rana cancarivora

B. Purpose
Different result cells smear vagina. Determined cycle phase happened of
female animal

C. Benefits
Can be know the induction ovulation of Rana cancarivora

D. Work Procedure
1. Lifting the pituitary gland
a. Entered the scissors in the corner of the female frog jaw, cut behind the
posteromedial eye, then passes head to the occipital region and finally to
the other jaw so that lifting the head.
b. Flip the skull and look for broad expanse formed by the bones at the base
of the cranium. Pituitary gland is located just behind the optic chiasm.
c. Entered sharp scissors into the brain cavity, and cut through the bone
toward the anterior skull base, to avoid injury to the brain tissue. By
using small tweezers, basic behind the cranium and look for pituitary
gland orange and shaped like a kidney.
d. Place the pituitary gland in a petri dish containing distilled water 2cc, and
do a few minutes later injection.
2. Injection
a. Hold the female frog firmly on his feet, do injection on posteriolateral
cavity. Avoid injury to the skin veins, vein ventral abdominal cavity and
other vital.
b. Females that have been injected, and then placed in a plastic vessel
containing water at a depth of one inch.
3. Stripping
Take a frog that has been stored for 24 hours, then do massage stripping
slowly, and bend forward towards the pelvis.
Then apply pressure on the front of the stomach backward.

F. Discussion
In this lab we will observe the induction of ovulation in the frog that is done
with three phase, that is:
1. The first stage is the removal of the pituitary gland. In observation of the
pituitary gland which, the gland is white, with a shape such as the kidney
and its size is very small. The glands under the cranium, which is then
shredded by adding distilled water to be easily dissolved, then put in a
centrifuge, then will appear the two layers are separated from the sediment
contained in the bottom of the tube and not used again, while there is a clear
liquid on the first layer is taken and then injected the female frog. Pituitary
gland located just behind the optic chiasm, cream-colored and shaped like a
kidney. The mechanism of ovulation is influenced by hormonal, neural, and
the periodicity of light. Ovulation in the frog oocyte release Occurs after the
first polar body, walls and follicle theca externa cells of follicle rupture.
Follicle growth due to the influence of the hormone FSH (folicle stimulating
hormone) produced by the anterior pituitary gland, the follicle cells are able
to produce the hormones estrogen and progesterone.
2. The next stage is the stage in which the injection of the abdomen of the
female frog pituatari injected fluid that has been put into a centrifuge, then
the frogs left in the pond that has been filled with water, then the frogs were
left for 24 hours, after that we go on The next phase that is stripping.
G. Conclussion
As can be concluded in this lab is to obtain eggs and fertilization process at the
desired time in large numbers, can be used suspension pituitary gland
hormones, with regard to the environmental conditions of female frogs that
have been injected suspension pituitary gland.
