THE ONION LAB.

Observation of onion cells.

Hypothesis: .

Materials: We will need

Tweezers, onion, methylene blue, eyedropper, Petri dishes, microscope, water, bottle of
water, slides and covering slides

Description:

1.- Take an small portion of the onion and with the help of the tweezers, take the transparent
layer that is between two layers. Put the onion skin on the slide.

2.- Now, add a drop of methylene blue to the skin and wait 2 minutes.

3.- Now we have to wash the methylene blue excess. Hold the onion and the slide with your
tweezers, put the slide in the dish and tilt the slide forming a slope. Now, let some water run
on the slide carrying away the blue ink while holding the onion skin.

4.- Once the skin is washed up, dry the bottom of the slide, cover the skin with the cover slide
(very carefully) and put it on the stage of the microscope.

5.- Focus the microscope using the smallest objective and observe. Draw what you see.

6.- Now, turn the nosepiece till you use the second objective. Observe again.

7.- At the end of the activity, wash everything up, throw the onion away and leave the lab in
perfect conditions.

3.- What have you learned or discovered out of the activity?

4.- Write a conclusion matching your hypothesis and explaining how it has proved your
hypothesis or not.
