The difference m quality of sedation induced by the new non-hypnotic CNS depressants can be readily distinguished from that induced by subhypnotic doses of the classical CNS depre_;sants by suitable tests in ,nice. The degree of depression ,observed m mice isolated from each other is compared _o the depressmn produced when mice are grou-. ped together (measured by the photocell cour.'_er technic of D_wsL Doses o,* i;e classical hypnotic agents which depress isolat,_:d :,pontaneous acrivitv of grouped mice are many rimes zhose required to depress isolated mice, where,. as doses of the new non-hypnotic depressants which sup-
press group spontaneous activity and which depress indi-
vidual activity are similar. The mild depressant action of the newer agents was also quantified by measuring effects on hexobarbital hypnosis in mice, and both the direct photocell measurement and the indirect hexobarbital method are employed for study of LSD antagonism. Reserpine causes marked reduction of group sponta- neous activity, and also prolongs hexobarbital sleeping time. The former action is not affected by LSD whereas the latter action is considerably reduced. The proposed mediator of Reserpine action, serotonin, also depresses spontaneous activity and prolongs hexobarbitai hypnosis, and both activities are antagmfized by LSI). The amine oxidase inhibitor Marsilid causes a mild depression of spontaneous activity which is converted into excessive stimulation by LSD, but its extreme effect in prolonging hexobarbital hypnosis is not affected by LSD. The depressant activities of Frenquet (azacyclonol) in both tests are, however, actually increased in the presence of LSD. Theoretical considerations of these interactions will be discussed. w8 (,E io'/+8o) NKT/ev 1256 ;_Dw
