B. Base theory

1.Cork tissue

Cork tissue is a tissue composed of parenchymal cells of cork. In dicotyledonous


plants. A tissue formed by cambium cork , cork felogen and is adjacent to or
beneath the epidermal tissue. Cork tissue formed in the direction in which is
called feloderm living cells, whereas cork cells are formed outward is called
felem and dead cells the cell form a box, having cell wall thickening by suberin.
And are impermeable (not translucent water)

2. Plasmolisis
Plasmolisis is a incident that plasma membrane free for cells wall because liquid
hipertonis.

If a plants cell is placed in concentrated salt solution ( hypertonic) . plants cell
lose water and turgor pressure , resulting in weak plants cell. Plants with cells in this
condition wilt. Lose more water will cause plasmolisis : keep the pressure decreases until
at a point where the protoplasmof cells scraped
from the cell wall. Causing the distance between
cell walls and membranes. Finally cytorrhysis – the
collapse of the entire cell wall-can occur. There is
no mechanism in plants cells to prevent excessive
water loss, also get water in excess, but
plasmolysis can be reserved if the cell is placed in
hypotonic solution . same processes in animal cells
is called crenation . fuild in the cells of animals out
because of diffusion.

Plasmolysis occurs only in extreme conditions , and rarely occurs in nature. Usually occur
accidentally in the laboratory by placing the cell at high salininity solutions or sugar
solution to cause ekosmosis , often using elodea plants or epidermal cells onions that
have color pigment so that the process can be clearly observed.

3. Osmosis
Osmosis is the diffusion of water solvent from hypotonic to hypertonic solution
by passing through semipermeable membrane. Every living organism has semy
permeable membrane system ,
Therefore, its possible if osmosis always occurs in life system of organism.

E. Procedures
1. Observing the cell of cork
 Make section of cork from available plant by using a razor. Try to cut as thin as possible.
 Put the section at the object glass
 Put the object glass on the microscope
 Watch cell of cork
 Draw the structure of cork
2. PLASMOLISIS
 Make section of beneath epidermis of Rhoeo Discolor leaf
 Put the section at the object glass
 Put the object glass on the microscope
 Watch the epidermis tissue
 Draw the observing result before dropped by 10% salt liquid
 Drop the section with 10% salt on the preparat
 Draw the observing result before dropped by 10% salt liquid
