MicrotoxOmni Software version 4.

The MicrotoxOmni Software has been upgraded by Modern Water Inc to allow Microtox users to easily run tests,
visualize data and associated calculated statistics and generate reports. Upgrades on v4.2 include:

1. New Operating Systems Compatibility: New version has been completely rewritten using the latest software
development tools, and is now able to load on state of the art operating systems, including Windows 7 64-bit Systems

2. Streamlined User Interface: The user interface has been simplified to streamline task flow for repeated use of the
same methods, including Test Protocol Selection sorted by frequency of use

3. Easier access to QA procedures

4. Easy to create custom protocol

5. Test reports can now be converted to pdf format

6. More Intuitive Test Selection Assistant

7. New Test Tutor Format, with more information at your fingertips

8. Improved Database Handling: The result database structure has been simplified, so that all data resides in the
database of your choice and can be sorted using the filtering functions

9. Trend Monitoring Updated

10. WET Method Compatible: Upgraded data quality and regression calculations to mirror WET Method

11. New Methods Available:

a. Confirmation Test for presumptive positives

b. 2% Screening Test for suspected high toxicity samples

13. More flexibility within both the ISO and Zinc Sulfate Standard Tests. In addition, a new protocol that reagent is
released on has been added to the Zinc Sulfate test that allows for better reproducibility and higher accuracy.

Calculations
The Calculations tab is the first tab on the MicrotoxOmni Settings dialog box. This tab provides the User with the ability
to modify the way calculations are performed and the manner in which data is displayed.

 The 'Use EC' and 'Use IC' option buttons allow the User to determine whether EC or IC is used throughout the text
of the application. The Default Value selection box, set to 50, allows the choice of the actual value to be calculated,
i.e. EC50, EC20, IC25 etc., and will accept any integer value between 1 and 99.
 'Auto Calc' - when enabled, causes the system to find the best set of data from the points available to give the
smallest confidence factor when performing the regression calculations. Points which are used in the calculation
will be marked with the '#' symbol, whilst those that are unused are not marked.
 'Allow deletion of data points' - when enabled, this option allows the user to delete data points from the statistical
calculations, by selecting the data point in question from the data grid or graph, then selecting the Delete data
point option from the 'Data' menu. The calculations will be re-done and the deleted point will be ignored. Deleted
points are marked with the 'D' symbol.
 'Calculate toxicity units' - when enabled, causes the EC, IC concentration to be displayed in toxicity units on the
'Statistics' frame.
 'Use reverse regression' - when enabled, causes the statistical calculations to be performed using regression of
log Gamma on log Concentration (ASTM method), as opposed to regression of log Concentration on log Gamma,
which is initially used as default.
 'Plot a line of best fit' - when enabled, causes the line of best fit to be drawn for the currently selected set of data.

Once the 'OK' button is pressed the new options will be saved to the application Configuration File and these will apply
to all new windows opened thereafter.

Canceling A Test

Users can cancel a test at any time by clicking on the drop down arrow at the top of the well display dialog box and
selecting 'cancel test'. This will bring up a pop up box to confirm the request to cancel the test. The Well Display and
Test Tutor (if opened) window(s) will then close. Data from the test will not be saved and the test may not be resumed
at a later time.

Change Passwords

This dialog box allows the MANAGER or USER to change and verify their own passwords.

Color Correction

The color correction procedure used by the MicrotoxOmni v4.2 is improved from the previous version. Absorbance
readings are required for each sample dilution, and the results can be entered using the following procedure: In the
Sample Test Results window, click the drop down arrow next to the Color Correction Mode icon and highlight 'Manual'.
An extra column is added to the data grid for all rows except the Control data row. Select the cell corresponding to the
sample dilution for which you wish to enter the absorbance data, and then simply type in the reading. On pressing the
<ENTER> key, the color correction is performed and the new gamma and percent effect values are calculated.

MicrotoxOmni Software version 4.2

Create a Custom Protocol

To create a Custom Protocol click File on the Menu Bar and select 'New Custom Protocol'. The Custom Test Protocol
dialog box will open. This dialog box has a section for naming the new custom protocol, this is the first step in creating
a new custom protocol. The dialog box also allows the User to start with 1 of the 17 Standard Protocols or use the
Test Selection Assistant to suggest a test protocol with which to start.

After a Test Protocol is selected, the User then modifies the parameters on the 4 tabs (within limits) clicks 'OK' and the
Custom Protocol is saved with the assigned name and ready for use. The new Custom Protocol can be opened from
the 'Run Acute Toxicity Test' tab on the main screen or from the Menu Bar → File→ 'Run Acute Toxicity Test', selecting
'Custom Protocols' and clicking on the appropriate custom protocol.

Please note: Test Tutor does not function with Custom Protocols.

Delete Custom Protocol

To Delete a Custom Protocol click on File in the Menu Bar and select 'Delete Custom Protocol'. Then highlight the
name of the Custom Test Protocol you want to delete and click on 'Delete'. A delete confirmation splash screen will
appear, click 'Yes' if you really want to delete the highlighted custom protocol, then click close if you are finishing
deleting custom protocols.

Delete Sample Results

This function allows for deletion of Sample Results, MANAGER level access is required. Clicking on Delete Sample
Results open a list of all the sample results in the current database. Click on the Sample Result you wish to delete and
then click the 'Delete' button at the bottom of the window.

Displaying Test Protocol Parameters

When the Test Protocol Parameters dialog box is opened the User will see the main dialog box with the name of the
Test Protocol, a Sample description section and 4 tabs for: Test Setup; Dilutions; Times and Advanced.

In edit mode the Sample Description box allows the User to enter a brief description to identify the current sample.

The 'Test Setup' tab displays control and sample parameters such as number of controls and samples plus control and
sample replicates.
The 'Dilutions' tab displays the number of dilutions, initial concentration, Dilution factor and concentration units.

The 'Times' tab displays the read time(s).

The 'Advanced' tab has a section for Reagent Lot number input if running a QA Test. The 'Advanced' tab also has a
'Keyboard Entry' check box. The 'Keyboard Entry' check box, when enabled, allows the User to type readings in at the
keyboard. When disabled, the system will attempt to take readings directly from the instrument.

In edit when creating a Custom Protocol or setting up a Site/QA Trend Monitoring the User may change the values of
certain parameters within valid limits, for example, setting the number of sample dilutions to values greater than the
instrument is capable of holding will cause an error message to be displayed when the 'OK' button is pressed.

Editing Data

Data which has been stored in the application database may be edited by Users who have an access level of
MANAGER, and this is achieved from the Sample Results Window by simply clicking on the cell of the appropriate data
grid representing the value you wish to change, and then re-typing the desired value. Note that only cells in columns
that represent light readings may be edited (i.e. the Io and It columns), other columns, such as 'Gamma' or '% effect'
cannot have their values altered. When data is edited by Users of MANAGER access level, the new value is displayed
on the data grid in brackets, {} to highlight the changed cells. Even if the values are changed back to the original
readings the brackets remain, indicating that the values were changed at some time in the past.

Exporting Data

Sample Results data may saved in an Excel format by selecting the 'Export Data' icon from the Sample Results window.
The data is then exported to the file/folder of your choice.

File Menu

The File Menu has options for: Open Test Protocol - allows viewing of the various tests protocol parameters; New
Custom Protocol - allows creating a custom protocol; Delete Custom Protocol - for deleting previously created custom
protocols; Delete Sample Results - for deleting sample results; Manage Trend Sites - for managing QA and Trend
sites; Log Off - to log off and allow another user access to the application; and Exit - to exit the application.

Instrument

The Instrument tab allows the User to adjust the communications settings for their PC, allowing connection of the PC
to the Microtox 500 by whichever Comm port is required etc. The main option is the Comm Port list box, which the User
can set to the correct port from which the instrument is connected. On pressing the 'OK' button, the current settings
are saved to the applications Configuration file and are used as default thereafter. Pressing 'Cancel' causes any
changes made to be ignored.

The Instrument Test option allows the User to test the communications between the PC and the Microtox 500 by taking
a single reading from the instrument. The User may select the operating mode of the instrument and then click start to
request the reading. Then the value obtained is displayed. If no data is received then there may be problems with the
communications link between the PC and the instrument, and therefore it is advisable to check the physical connections
as well as ensure that the PC's Comm port has been correctly set up.

Manage Trend Sites

A sampling site may be added to the database to allow the data obtained from that site to be used for Trend Monitoring
by selecting 'Manage Trend Sites' from the 'File' menu. This option will only be available when the User has logged on
to the database and has an access level of MANAGER. If this option is selected and no previous site or QA test details
have been recorded in the database, then the User will be prompted to select a file name for the test template file which
will be created to store test parameters for the tests to be used for trend monitoring. The dialog also contains a Start
with Test Protocol section, from which the User may select a test protocol from 11 standard protocols as the starting
point for creating the specified test for this site.

Note: Screening Test protocols cannot be used for Trend Site Monitoring.

After creating the Trend Site Test Protocol, press 'Add' and these parameters are stored to the specified trend
monitoring template file, and may no longer be edited. The new site data will then be added to the database. Trend
tests for this site may then be run by selecting 'Trend Monitoring' from the 'Test' menu as usual, choosing the desired
site from the displayed list.

The Trend Site Management dialog will also display a list of any sites currently stored in the database.

Reports

The Reports tab allows the User to set up the overall layout of MicrotoxOmni reports. The User can create the title of
the report and also add a picture from a file, such as a bitmap image, by checking 'Show Custom Picture' and specifying
the file name clicking the 'Browse' box and selecting the appropriate file. The image can then be positioned on the
report as required.

The User can then add 'Custom Text' for the report by typing in their preferred text in the Custom Text box. Reports
can be further customized by selecting any of the 4 display options: Plot of Gamma, Plot of % Effect, Signature Line
and Time/Date.

This allows the User to select which items they wish to display or remove from the report, by checking or unchecking
the various options provided.

Select a Test Protocol

Selecting a test protocol for running a test can either be done from the main screen by clicking on 'Run Acute Toxicity
Test' or 'Run Specialty Test' or it can be done by clicking 'Test' on the Menu Bar and scrolling down to the two previously
mentioned Run Test options. Both the main screen and Menu Bar 'Test' options step you through the same series of
dialogue boxes for selecting a test protocol.

Clicking on 'Run Acute Toxicity Test' takes you to the Select Test Protocol Dialog Box. This Dialog Box presents you
with 3 main options: selecting from any of the 17 Standard Protocols; selecting from previously created Custom
Protocols; or selecting from previously created Site/QA Tests.

There is a pop up on the Select Test Protocol Dialog Box screen for Test Selection Assistant. See Test Selection
Assistant for additional details.

The 17 Standard Protocols in MicrotoxOmni v4.2 are:

 2% Basic Test
 2% Screening Test

Select a Test Protocol

Selecting a test protocol for running a test can either be done from the main screen by clicking on 'Run Acute Toxicity
Test' or 'Run Specialty Test' or it can be done by clicking 'Test' on the Menu Bar and scrolling down to the two previously
mentioned Run Test options. Both the main screen and Menu Bar 'Test' options step you through the same series of
dialogue boxes for selecting a test protocol.

Clicking on 'Run Acute Toxicity Test' takes you to the Select Test Protocol Dialog Box. This Dialog Box presents you
with 3 main options: selecting from any of the 17 Standard Protocols; selecting from previously created Custom
Protocols; or selecting from previously created Site/QA Tests.

The 17 Standard Protocols in MicrotoxOmni v4.2 are:

Clicking on 'Run Specialty Test' gives you the option of selecting protocols for either Quality
Assurance or Genotoxicity.

For Quality Assurance testing you can select either of the 2 standard tests:

 Phenol Standard
 Zinc Sulfate Standard

use the drop down arrow and select the standard you want to run and click on Quality Assurance to start the test.

The Genotoxicity option is for running the Mutatox (Direct) Medium Test.

 Running Test Protocols

To run a Test Protocol, select 'Run Acute Toxicity Test' or 'Run Specialty Test' on the main screen or select Test on
the Menu bar and then select either of the 2 previously mentioned Test options.
Selecting 'Run Acute Toxicity Test' opens the 'Select Test Protocol' dialog box. There are 3 primary options in this
dialog box; Standard Protocols - allowing selection of 1 of the 17 Standard Protocols; Custom Protocols - allowing
selection of previously created custom protocols; and Site Tests - allowing selection of previously created site specific
test protocols. You also have the option of choosing the Test Selection Assistant to guide you through the test selection
by clicking on 'Assistant' at the bottom of the window.

Selecting 'Run Specialty Test' opens the 'Select Specialty Test' window for selecting either of the 2 Quality Assurance
test protocols or selecting 'Genotoxicity' for the Mutatox (Direct) Medium Test. To run a Quality Assurance Test, click
on the drop down arrow under the 'Quality Assurance' box to bring up both QA tests, then highlight the QA test you
wish to run and click the 'Quality Assurance' bar. This opens the Test Protocol Parameters dialog box. You will need
to enter the Reagent Lot number on the 'Advanced' tab and then click 'OK' to begin the test.

On pressing 'OK' the 'Parameters' dialog is closed and a Sample Information window opens for you to input detail on
the sample; Location and Sample Name. Clicking 'OK' closes the Sample Information window and the Well Display
window will now open. If the Test Tutor system has been enabled, then the Test Tutor window will also open and will
guide the User through the full test protocol from preparation of samples to running the test, otherwise the User is
prompted to press the 'START' button on the Well Display to begin the test. The User is then guided through the
acquisition of light readings by the message labels on the graphic display.

On completion of the test, statistical calculations will be performed on the newly acquired data and the results displayed
in the Sample Results window. If necessary, color correction data may also be entered for the test sample after the
test is complete (see Color Correction).

 Sample Results

Sample results are displayed immediately after a test is completed and saved or by selecting Sample Results in the
Test Menu for viewing previously saved results. Using Sample Results in the Test Menu brings up a list of all sample
results in the current database. The list can then be filtered by: Test Protocol; User; From/To Dates; and Location.
Specific results are then selected by clicking on the appropriate row and then clicking the 'Open" button at the bottom
of the screen.

The icons at the top of the window allow the User to: View the Test Parameters; select the Color Correction Mode
(None or Manual); Delete Selected Row, the Control row cannot be deleted; Create Report and Export Data.
The Sample Results window consists of two sections which display a graph and the data grid, as well as a Statistics
frame which displays the calculated statistical data for the currently selected set of results. The Graph section has a
display option allowing the User to change the Plot Y-Axis via selecting either Gamma or % Effect (default). The Data
Grid section displays the individual sets of test data taken for a single sample at each test time simultaneously. Each
tabbed panel is labeled with the time after which the particular set of readings were taken. The readings, calculated
gamma and % effect values, or any other relevant data values for the particular type of test being examined are
displayed in a data grid, and the statistical values (if available) for the currently selected grid are displayed in the frame
at the bottom of the form.

The line of best fit for this set of data is also drawn on the gamma versus. concentration plot.

The User may select a row of the data grid by moving the mouse pointer to the left hand edge of the selected row.
Clicking the left mouse button will cause the row to be highlighted, and this will allow the user to delete that point from
the calculations (see Deletion of Data Points) by clicking on the 'X' icon,or restore a point that has previously been
deleted by again clicking on the 'X' icon.

The 'Statistics' frame will display calculated values depending upon the type of test run and the set of data currently
selected. Where appropriate, the calculated EC/IC concentration and it's 95% confidence limits are shown, the 95%
confidence factor, the estimating equation used in the linear regression calculations, the slope of the line of best fit, the
coefficient of determination and the correction factor used to adjust timed sample readings. If the User wishes the
EC/IC concentration to be shown using Toxicity units this needs to be done via the Tools Menu → Options and then
checking the Toxicity Units box on the MicrotoxOmni Settings Window.

Additional results specific to particular test are also displayed in the Statistics frame.

Test data stored in the application database may be edited by Users with MANAGER level access privileges (see
Editing test data). The values for light readings only may be edited, and a full history of edits performed on test data is
maintained i

Specifying a QA Trend Test

To specify a QA Trend Test, from the main Menu bar, select Test → Trend Monitoring, click on QA Test and then
highlight the standard you wish to monitor.

Trend Monitoring

The Trend Monitoring Setup dialog box provides the ability for Users to monitor the results of tests performed at
specified test sites or for QA tests over time, allowing any trends in the results to be identified. For trend monitoring to
be performed, a test protocol must be selected to be used to obtain the trend data for a site or QA test. The User must
first create a new trend site via the 'Trend Sites Management' dialog box, available from File Menu → Manage Trend
Sites. The created trend site is then stored in the application database, and the test parameters associated with each
site test are stored in a separate test template file. Site or QA tests can then be run by any User as an option from the
'Run Acute Toxicity Test' button on the main screen or from the Test Menu. The results can then be plotted in the Trend
Monitoring window, which shows how a pre-selected value, such as EC50, varies over time, and can be filtered to
show data obtained by a specific User. To view the trend monitored data, the User must be logged on to the database
with an access level of MANAGER. The 'Trend Monitoring' option can then be selected from the 'Test' menu, which
will open the 'Trend Monitoring' dialog, and allows the User to decide whether to trend data for a specific site or QA
test, which value is to be monitored, and whether to use data only for a single User. All data or a specified date range
can be plotted. The Trend Monitoring data is then read from the database and plotted on the Trend Monitoring window.

Trend Monitoring Graph Options

Trend Monitoring Graph Options allows the User to alter the way the Trend Monitoring graph is displayed, and is
opened by selecting Test → Trend Monitoring, selecting the Site/QA Test to be monitored and then clicking on the
'Graph Options' tab in the Trend Monitoring window.

The 'Points' tab allows the User to configure the number of data points to be displayed on the graph at one time, to
allow a clearer display of results.

The 'Limits' tab allows adjustment of the way the SPC calculations are performed. The three option buttons allow the
choice of using 'rolling' control limits (where the control limits are recalculated for each data point), 'fixed' limits (where
the values of the limits after the specified number of control points are used for each point thereafter, producing straight
lines for the upper and lower limits) or switching off the limit lines completely. The 'Number of Points (used to calculate
limits)' box allows the choice between 10, 15 and 20 points before the SPC limit lines are plotted.

The 'Standard Deviations' box determines how many standard deviations away from the mean the control limits are
calculated.

The '(Apply) SPC Rules' check boxes allow the User to select which of the following three SPC rules should be flagged
if broken by a point on the monitor graph.

 Rule 1 - Any point above the upper control limit or below the lower control limit.
 Rule 2 - Any set of 5 sequential points that are all above or all below the mean.
 Rule 3 - Any set of 5 sequential points that are all rising or all falling.

The 'Y-Axis' tab allows the User to choose whether to allow the graph to automatically resize to fit the range of
monitored values for the data currently displayed, or to select and specify their own fixed maximum and minimum Y-
axis values.
Pressing 'OK' will close the Graph Options dialog and any changes made to the options will be enforced on the graph,
whereas pressing 'Cancel' will cause all changes to be ignored.

Trend Monitoring Window

The Trend Monitoring window consists of a graph displaying the selected monitor value (i.e. EC50) against Time for
the chosen Site or QA test, along with a table showing the actual data values plotted and the User who performed each
individual test. The window is opened after the User has selected the monitoring options on the Trend Monitoring Setup
dialog box.

The Trend Setup tab re-opens the Trend Monitoring Setup dialog, to allow the User to choose a different Site/QA test,
change the monitored value, etc., and Filter Results.

The Graph Options tab - opens the Trend Monitoring Options dialog, which allows the User to alter the way the graph
is displayed.

The graph displays the data as a series of linked points, with the monitored value on the y-axis and the date and time
of the particular test on the x-axis. Note however that the x-axis is not scaled according to time, each data point is
plotted at a constant distance from the previous one. If the data is not being monitored specifically for a single User,
then the cumulative mean values of the data may also be plotted as a series of linked points after the specified number
of control points has been reached. Also the SPC control limits may be plotted, calculated using the desired number of
standard deviations from the mean (as determined in the Graph Options dialog 'Limits' tab). If any points break one of
the SPC rules (which can again be set using the Graph Options dialog), then the point will be marked with the number
of the rule broken.

The scale of the y-axis, the style of the control limits and other options may be changed using the Graph Options dialog.

The data grid displays the monitored value, date & time, and Use name for each appropriate set of data for the chosen
Site/QA test. The 'Users' frame contains a list of all the Users who performed tests to obtain the data for the selected
Site/QA test.

The Trend Monitoring window also contains 2 icons: 'Create Report', see MicrotoxOmni Report Viewer and 'Export
Data', see Exporting Data for additional details.

Close - closes the Trend Monitoring Window.
