Spectrophotometric Determination of Permanganate Concentration

Objective: To illustrate the measurement of concentration of unknown permanganate

sample using Spectrometry in the visible region.

Apparatus/Chemicals/Equipment:

 8pcs 100mL volumetric flask

 Cuvette
 2.00 mL Pipette

Chemical /Reagents:

0.02 M KMnO4

Procedure:

(a) Permanganate Standards

Prepare and standardize a 0.01 M solution of KMnO4. Then using a measuring pipette,
place 0.30, 0.50, 1.00, 1.50, 2.00, 3.00 and 4.00-mL portions into five 100-mL
volumetric flasks numbered 1 to 5, these will serve as your standard solution. Prepare
another 100 mL flask and place 1.30 mL of 0.01 M KMnO4, this will served as your
unknown solution. Dilute solution in each flask to the mark. Calculate the
concentration (ppm) of resulting standard solutions.

(b) Determination of Absorption Maxima

Using one of the permanganate solutions of intermediate concentration, measure the

absorbance over the wavelength interval 400 to 600 nm using distilled water as
reference. Read the absorbance at 20-nm intervals except in the vicinity of the
maxima and minima in the spectrum, where the readings should be taken every 5 nm.
Plot absorbance vs. wavelength.

(c) Determination of Calibration Curve

Select the wavelength of maximum absorbance. Measure the absorbance of the
standard solutions at this wavelength. Plot absorbance vs. concentration for each ion
and calculate the equation of the line using least square method.

(d) Analysis of the Unknown

Measure the absorbance of the unknown at the chosen wavelengths. Calculate the
concentration of MnO4- in the unknown.

(e) Determination of Limit of detection

Measure the absorbance of the blank ( or a very dilute KMnO 4 solution) 10 times.
Calculate the standard deviation of the blank.
