EL HONGO Gibberellafujikuroi
BIOSÍNTESIS DE GIBERELINAS
Las giberelinas" son terpenoides, compuestos que derivan del
difosfato de isopentenilo. Esta molécula se puede sintetizar por dos rutas
biosintéticas diferentes. La primera usa como intermediarios el
hidroximetilglutaril-CoA y el mevalonato (Fig. 1); la segunda,
descubierta más recientemente, usa como intermediario el fosfato de
gliceraldehído y el piruvato (Rohmer et al., 1993). Los experimentos de
alimentación con mevalonato radioactiva indican que las giberelinas se
sintetizan en Gibberella y en las plantas por la vía del mevalonato (Birch
et al., 1958; Coolbaugh, 1983; Domenech et al., 1996).

TRANSFORMACIÓN DE Gibberella
La identif!caéión y caracterización de los genes involucrados en el
metabolismo secundario de Gibberella requiere disponer de sistemas
eficientes de introducción de ADN exógeno y de mutagénesis dirigida. La
transformación de Gibberella se llevó a cabo por primera vez
introduciendo el gen de la nitrato reductasa de Aspergillus niger en
mutantes de Gibberella carentes de esta actividad enzimática (Sánchez-
Fernández et al., 1991). Posteriormente se han utilizado otros marcadores,
como son el gen de la transcarbamilasa de la ornitina de Aspergillus
nidulans, o un gen bacteriano de resistencia a higromicina (Leslie y
Dickman, 1991; Brückner et al., 1992; Fernández-Martín, 1998).
La transformación de Gibberella es integrativa y los transformantes
obtenidos muestran un elevado grado de estabilidad (Brückner et al.,
1992). Las frecuencias de transformación obtenidas son bajas (menos de
10 transformantes por microgramo de ADN), por lo que no es práctico su
empleo para clonación de genes por complementación. Una variante del
método de transformación por resistencia a higromicina permite que la
mayoría de los transformantes hayan integrado el vector por
recombinación homóloga (Fernández-Martín, 1998). El método permite
realizar experimentos de disrupción génica por integración dentro de un
gen o mutagénesis dirigida por integración y excisión del vector.
