Autoclave is a important
apparatus used for the
sterilization of any given
object. In appearance,
working and construction it is
similar as pressure cooker.
The object which is placed
inside the autoclave face a
high steam pressure and heat
thus the protein of
microorganism present in the
object get precipitated and the
object got sterilized. The
principle of autoclave is
based on this. This is not
suitable for the hygroscopic
substance, but suitable for
those which may get
damaged by dry heat

 Handle of Autoclave
This is simplest unit in
autoclave. As like other
handle in any apparatus
Handle is use to hold the
cover lid of autoclave
while placing and
removing object from
autoclave assembly. The
top of the handle is
made of thermo resistant
plastic which keep the
handle cool. Although
handle does not seek
that much attention but
one thing is to be keep
in mind- while removing
handle never place it on
the flower downside.

 Pressure Gauge (Meter)
This part of autoclave is
called pressure gauge or
pressure meter. The main
function of this is to
indicate the pressure
build-up inside the
chamber of autoclave.
The autoclave machine is
based on the pressure so
this unit become more
important. The specific
pressure should be
maintain with its
respective time to
achieve a sterilization. To
assure the safety of
autoclave and operator
the working condition of
pressure unit must be
checked periodically.

 Autoclave closing screw

As there is clip to close

the pressure cooker at
home there is same unit
in autoclave called-
Closing screw or simply
screw. All screw should
be tightly close before
using the autoclave. The
right procedure to close
these is that- first of all
place all screw in their
respective position and
then start fighting all of
them. If any screw
remain loose there is
possibility for vapour to
leek out. While opening
back make sure that the

vapours are condensed.
 Pressure Releasing unit- Whistle
As we have seen the
whistle in pressure
cooker, this is the same
in principle. Autoclave
working is based on the
pressure and when the
autoclave is heated
water start to convert
into a vapour and if this
process is continue then
there will be chance of
blasting, and the
pressure may increase
beyond the desirable
limit. This whistle
control the pressure by
releasing certain
amount of vapour by
lifting up itself.

 Safety Valve of Autoclave
Safety valve is one of the
important unit of
autoclave for the safety
point of view. Safety
valve is having a thin
layer of rubber and when
the pressure increase in
uncountable manner and
of the autoclave fails to
perform it’s action there is
a chances of explosion of
autoclave. To avoid these
dangerous situation there
is a safety valve which get
burst when pressure could
not resealed from basils.
Safety valve may choked
so regular inspection is
advisable.

 Manual Pressure Release Screw
This unit is called
manual release screw or
simply release screw.
When there is need to
release the remaining
vapour after completion
of sterilization cycle, this
screw may be open and
then object may be
removed from the
autoclave. All this
process should be carried
out inside of the aseptic
chamber to avoid any
type of further
contamination. This
screw should only be
open after the
completion of cycle.

 Heating Coil and Tray Stand
After adding sufficient
volume of water, heat
is supplied by this
heater so that the water
may convert into
vapour. The stand in
this picture is used for
two main purpose- (1)
To avoid the
swimming of tray and
(2) To indicate the
level of water that
should be filled inside
the autoclave. The
volume of water is
important parameter, if
water is exceed the tray
will float above this
stand and if water is
less the coil will burn.

 Object Placement Tray
The object showing in
the display is called tray.
Tray is used to hold or
place the objects which
are about to sterilized.
The tray is placed over
the stand and below that
was filled. The objects
like- glass ware and
media should be
wrapped in brown or
white paper before
keeping in the tray. Tray
are basically made of
either aluminium or
stainless steel. Object
should be placed at a
optimum distance to
each other.
