THE EFFECT OF HUMAN ADIPOSE-DERIVED STEM CELLS (ADSC) ON THE

FIBROSIS GRADING ON THE BLADDER OBSTRUCTION MODEL OF WISTAR


RATS

ABSTRACT

Introduction: Bladder Outlet Obstruction (BOO) was caused by a series of histological and
biochemical changes in the bladder wall, through the inflammation process in the bladder wall,
hypertrophy and fibrosis at the decompensation stage. The inflammatory process is characterised
by an increase in the organisation of extracellular matrix (ECM) due to increased synthesis of type
I collagen, increased expression of TGFβ1, and increased inflammatory products such as collagen
and fibroblasts and their receptors in the stroma and epithelium. This increase causes smooth
muscle dysfunction and decreased bladder compliance. Adipose-Derived Stem Cells (ADSC)
express cell surface markers, genes, and cell differentiation potential which are common as
Mesenchymal Stem Cells (MSC) and have been used for the reconstruction of various tissues and
organs. ADSC has an important role in bladder regeneration.

Methods and Materials: This study was an experimental randomized study using male Wistar
rats which were monitored at 2 and 4 weeks to determine the effect of ADSC therapy on TGF-β1
levels, type I collagen and degree of fibrosis, and histopathological improvement of the bladder
wall compared to control.

Result: Rats were divided into 4 groups. In week 2 BOO group, 1 sample was moderate fibrosis,
1 sample given ADSC was mild fibrosis, 3 samples were severe fibrosis, 3 samples given ADSC
were moderate fibrosis. In week 4 of the BOO group, 2 samples were moderate fibrosis, 2 samples
given ADSC were mild fibrosis, 3 samples were severe fibrosis, 3 samples given ADSC were
moderate fibrosis. Based on the calculations results there are influences of ADSC on Fibrosis BOO
Week 2 and 4 which is statistically significant. After ADSC administration, there was a significant
decrease in the degree of fibrosis in the second and fourth week (p-value <0.05).

Conclusion: Administration of ADSC on bladder outlet obstruction in mice model can


significantly reduce the degree of fibrosis, assessed through histopathological examination
