Aldehyde Fuchsin Colour

DESIGN TO DEMONSTRATE Elastic fibers, Mast cell granules, Mucins,


β cells of the pancreatic islets, certain hypophyseal β granules and Pituitary glands.

REAGENTS & PREPARATIONS


Iodine Solution 200ml : Ready to use. Store at 2–6°C
Hypo Solution 200ml : Ready to use. Store at 2–6°C
Aldehyde Solution 200ml : Ready to use. Store at 2–6°C
Counter Solution 200ml : Ready to use. Store at 2–6°C
TISSUE SAMPLE
3-5µ paraffin sections of 10% neutral buffered formalin Or Bouin’s
Fixative.
STAINING PROCEDURES
1. Remove Paraffin through xylene, rehydrate through graded ethanol and bring
sections to distilled water just prior to staining.
2. Place into Iodine Solution for 5 minutes and rinse in distilled water.
3. Remove iodine with Hypo Solution for one minute. Wash in running tap water
for 30 seconds and rinse in distilled water.
4. Place in 70% alcohol for one minute.
5. Stain with Aldehyde Solution for 10-15 minutes.
6. Rinse in 95% alcohol to remove all excess staining and rinse in distilled water.
7. Counter stain with Counter Solution for 1 minute and rinse thoroughly in
distilled water.
8. Dehydrate up through graded alcohol to absolute alcohol.
9. Clear in xylene and mount with a resinous medium (medilines DPX
mountant).

Expected results
 Elastic fibers, Mast cells granules, Mucins – Purple to violet
 β Cells – Purple to violet
 Background – Green

Notes
1. Prior to place into Aldehyde Solution slide should be placed in 70% Alcohol (Step 5).
2. Elastin should be exposed to Aldehyde Solution for 10-15 min (Step 6).
3. β Cell should be exposed to Aldehyde Solution for 30-35 min (Step 6).
4. Once a satisfactory stain has been achieved, the time should remain constant. Any
change in fixation, processing or section thickness warrants reviewing the staining time.
5. Avoid microbial contamination of reagents; otherwise increase in nonspecific staining
may occur.
6. The user must validate any changes made to the factory-released procedure.
7. Store reagent packs in an upright position when not in use.
8. Keep reagent packs out of direct sunlight and away from heat-generating sources.
9. Wear appropriate Personal Protective Equipment to avoid contact with eyes and skin.
