HEMALAB o Mechanical Stage – moves along x & y –axis

o Mechanical stage adjustment knob
 Very impt optical instrument used to magnify minute organism/ specimen in o Condenser – gather beam of light and directs it to the specimen
question such that the details of its structure will be visible to the naked eye o Iris diaphragm – regulates amount of light w/c illuminates the objective
o Iris diaphragm aperture – moves left or right
 3 types: o Light source – gives correct brightness
1) SIMPLE – magnifying lens; single lens - most common: tungsten halogen light
2) COMPOUND – has 2 lenses (eyepiece/ocular and objectives); o Coarse adjustment knob – gives the general view/ outline of the field
- magnifies up to 1000x o Fine adjustment knob – gives detailed view of the structure of the specimen
- “ Brightfield Microscopy” o Light adjustment knob – controls the brightness of the light
3) ELECTRON – magnifies up to 1,000,000x o Switch

 Parts:  Cover w/ plastic when not in use to avoid accumulation of dust


o Eyepiece/ Oculars – 10x magnification (diameter of object being magnified)  Carry microscope w/ both hands; dominant hand on neck, the other on base
* x means diameter (if 10x, means object is magnified 10x)  Solvents used to clean the objectives of the microscope
- could either be Monocular or Binocular o Xylol alcohol/ Xylene - carcinogenic due to benzene
o Interpupillary control – determines distance between two oculars o Lense cleaner solution
o Body tube/ Optical tube – holds the eyepiece in place o 70% isopropanol
o Arm/ Neck – provides support; attachment site of revolving nosepiece
o Revolving nosepiece – holds objectives in place; allows easy rotation of
objectives
o Objectives
* Low Power Objective
- working distance = 16 mm
- magnification power = 10x; total magnification = 100x
* High Power Objective/ High-dry objective/ high & dry objective
- working distance = 4 mm
- magnification power = 40x; total magnification = 400x
 numerical aperture (ability of microscope to gather light) = 0.65
> resolution (ability of mic to focus finer details of specimen)
> increased resolution = increased quality
 optical tube length (distance bet ocular & objective) = 160 mm
* Oil Immersion Objective
- working distance = 1.8 mm
- magnification power = 100x; total magnification = 1000x
 Oil (classified acc to viscosity)
> type A – low viscosity (fluorescence & darkfield mic.)
> type B – high viscosity (brightfield & clinical mic.) = Cedar oil
> type C– very high viscosity (inclined mic./ long objectives)
