It would be my utmost pleasure to express


my sincere thanks to the Principle of school
Mrs. BHAWANA BHANOT for allowing
me to perform the experiment in Chemistry
Laboratory. I want to thank my chemistry
teacher, Mr. Bhupender Negi for guiding
me in the project. I am also thankful to the

Chauhan and Chemistry Lab Assistant
Ms. Sheetal Mishra for lending me a
helpful hand in this project. Their valuable
guidance, support and supervision all
through the project titled, “Extraction of
Determination of Caffeine Contens in
Various Tea Samples” are responsible for
attaining its present form.

AIM:- Extraction and Determination of
Caffeine in Tea Samples.

APPARATUS:- beaker, test tube,


flask, burner, filter paper, weighing
machine & separating funnel.

MATERIAL REQUIRED:- tea,


chloroform, distilled water, calcium
carbonate and hydrochloric acid.

THEORY:-
Use difference in the solubility of caffeine
and tannic acid in water and chloroform to
separate them from tea leaves. Caffeine is
much more soluble in chloroform than in
water. But tannic acid is extensively soluble
in water as well as chloroform.
PROCEDURE:-

Components from Tea Sample - Step I

1. Take about 100ml of distilled water in


250ml conical flask.
2. Add 6gm of tea and boil it for 4 to 5
minutes. Then cool it and filter.
3. Repeat this process again with 100ml of
distilled water and residual tea leaves.
4. The filtrate in each case should be
collected.
5. Now the wet tea samples may be dried in
an over and weighted.

Removal of Tannic acid from Filtrate –


Step- II

1. The filtrate obtained in the first step is


now boiled with 2-3 grams of calcium
carbonate to get tannic acid, present in tea
extract, precipitated. On leaving it for ten
minutes ppt. is calculated.
2. Filter the solution without disturbing the
ppt carefully. The ppt should be transferred
to filter paper only after all the filtrate is
removed.
3. Tannic acid has been extracted in the
form of calcium ternate which on treatment
with hydrochloric acid gives tannic acid.
4. The filtrate is now free from tannic acid
and can be used to extract caffeine.

Extraction of Caffeine from Tannic


acid free filtrate – Step III

1. Take tannic acid filtrate obtained in


step(II) and add 100 ml of chloroform in it
in a separating funnel and allow it to stand
for 10 minutes.
2. Again the same procedure as done in (I),
then mix it with the previous one.
3. From this chloroform extract, chloroform
may be distilled or evaporated.
4. The pale green residue obtained is crude
caffeine. Repeat this experiment with other
brands of tea and record your observation in
a tabular form.

 Weight of tea used = W1 gm.


Weight of Caffeine = W2 gm.
Calculations:-

Formula for percentage of


solute caffeine in different = W1 / W2 *100
brands of tea

1. for double diamond –


W1 = 6gm,
W2 = 0.8gm
Percentage of solute caffeine = 0.8/6 * 100 = 13.33%

2. for The Taj –


W1 = 6gm,
W2 = 0.7gm
Percentage of solute caffeine = 0.7/6 *100 = 11.6%

3. for Red Label –


W1 = 6gm,
W2 = 0.72gm
Percentage of solute caffeine = 0.72/6 *100 = 12%

Result:-
The sample of Double Diamond contains
highest amount of caffeine. Thus –
1. Gives better taste than others.
2. Is more harmful for our health.
Precautions:-

1. Chloroform should not be smell


directly as it is toxic.
2. The water used should be distilled.
3. Apparatus should be clean.
4. NO prolonged heating should be done
as water may evaporated in large
quantity along with volatile solute.
