Introduction of foreign DNA into plants
(a) The cloning vector is a plasmid carrying
sequences essential for replication and
selection in E. coli as well as Ti plasmid
sequences. Foreign DNA can be inserted
into this vector and ultimately integrated
into the genome of the host plant. In the
example shown, the luciferase gene from
firefly is transformed into a tobacco plant
using the Ti plasmid. (b) Watering the plant
with a solution of luciferin (the substrate for
firefly luciferase) results in the generation
of light by all plant tissues. This plasmid
(Ti), from the soil bacterium Agrobacterium
tumefaciens, can transform many kinds of
plants. The plasmid grows first in the
bacterium. Upon infection of the plant, the
bacterium passes the plasmid into the
plant cell. From the infected portion of the
plant, another plant can be grown in which
the plasmid has infected all cells (as in the
tobacco plant shown.) Offspring will also
have the plasmid in each cell.
