1.0 PURPOSE : The plethysmometer use for screening of

information or oedema in mouse or rat
The procedure is written to describe all steps for operating plethysmometer

2.0 SCOPE : The plethysmometer is use for neuropathic pain, hyperalgesia,

inflammation, joint pain in mouse or rat

3.0 RESPONSIBILITIES : Thecher in charge of laboratory as well as Thecher. Thecher

in charge of activity are responsible

4.0 DEFINITION : The digital water plethysmometer is designed to provide a highly

useful tool in the measurements of small volume changes this test is typically
used to follow the evolution of the inflammatory response experimental
induced in rodents and to screen properties of pharmacological substances

5.0 Material and equipment: button touch auto- calibration mode making hassle free
calibration
• Stable & comfortable reading on graphical LCD display
• Six point calibration 0.50 ml,1.00ml,2.00ml,3.00ml,4.00ml& 5.00ml

6.0 PROCEDURE: Firstly weigh all the rat and number them accordingly on their tail
o Make a mark on both the kind paw of the experimental rat from 2 cm above
the paw it’s done just to ensure the proper and same dipping of paw each
time
o Note down the initial paw volume of left and right paw of rat just by
observing the level of displaced Mercury
o Divide the animal into two group having minimum 04 rat
o In one group we will inject simple saline and in in another group we will
inject our drug of study that is indomethacin
o After 30 min inject 0.1 ml of carrageenan in the left paw of rat and it will
treated as control group
o The right paw will kept as it is because it will treated as reference for non
inflamed paw of comparison
o Note the paw volume of both legs of control and indemethacin treated rat
as 15,30,60 and 120 min after carrageenan

o Calculate the % difference in both paw of rat of control and indomethacin

treated group as per the reduction of oedema

7.0 PRECAUTION/SAFETY MEASURES: keep the instrument clean and dry

Do not handle the instrument with wet hands

8.0 Reporting: Result should be recorded in a record book if there are problem during
the test consult the laboratory in- charge
