GSK461364 is an ATP-competitive inhibitor of Plk1 and forms a rapidly reversible complex with Plk1. It has a 400-fold greater potency for Plk1 than for Plk2 and Plk3. It is > 100-fold selective with respect to PLk2, PLK3, and 48 other kinases tested. Treatment of 120 cell lines with GSK-461364 led to growth inhibition with an IC50 of < 100 nM in over 90% cases, with a net cytostatic response in about 40% and net cell death in 45% of cell lines tested.
