2. DNA, RNA, TRANSCRIPTION AND TRANSLATION Cells use the two-step process of transcription and translation to read each gene and produce the string of amino acids that make up a protein. In this lab and the associated assignment we will perform this process on paper. Please ensure that you are familiar with the exercise before you leave the lab, so that you can do the assignment. For example: The following represents a single (sense) strand of DNA

1. Write out the other sequence (anti-sense) needed to complete the double stranded DNA molecule (you can write on this paper). 2. When transcription starts the strands separate to allow RNA to make a copy. Write out the corresponding RNA copy that would be produced from the above DNA strand. 3. Examine the RNA strand and (using the universal genetic code table) look for the correct starting position for the translation process. This will correspond to the MET amino acid. Make sure you also identify a suitable end code (Ter = Stop). Work your way along the RNA strand using the chart to position the amino acids into the correct sequence to from a polypeptide. Write out the amino acid sequence. 4. Begin this process with one of the sense strands from the assignment, this is the actual assignment exercise, which you may complete in the lab if you have time, or complete afterwards.

The aim of this assignment is for students to: Become familiar with base pairing that compose DNA and RNA Understand transcription & translation Understand the triplet code Observe the effects of changes in the DNA sequence

Submit your assignment in electronic copy to the assignment drop-box in Moodle, or hard copy to the assignment drop-box on level 3, building 115. The work submitted must be your own work with no plagiarism. Marking schedule is available for each course assessment to assist you with assignment content.

Assignment 1 Transcription Translation You are provided with several pages of sequence data from the Human Genome. Select a pair of sequences (e.g. 1a and 1b) from this page then: 1. Write out the other sequences (anti-sense) needed to complete the double stranded DNA molecule, for both sequences in the pair. 2. When transcription starts the strands separate to allow RNA to make a copy. Write out the corresponding RNA base for each DNA base (sense strand) to make RNA strands for both sequences. 3. Examine the RNA strands and look for the correct starting position for the translation process. This will correspond to the Met amino acid (use the universal genetic code chart provided). Highlight an appropriate Start triplet on your sequence. Make sure you also identify and highlight a suitable end code (Ter/terminator). 4. Write out the amino acid sequences that forms polypeptides, between the start and stop triplets 5. Show how your amino acid sequence corresponds to your RNA sequence. 6. Continue the process until you have accounted for you entire original paragraphs of DNA sequences 7. Compare the 2 sequences in your pair and try to work out what is the change between the 2 sequences, and what effect this had on the peptides. NB- these sequences are available on Moodle if you wish to directly work on a computer. For those interested the following is the summary information from which these sequences were derived from GenBank (prior to being altered for this lab).
