PROFESSIONAL EXPERIENCE University of Maryland, Baltimore Postdoctoral fellow, Greenbaum Cancer Center, Dec 2009-Present. Studying the mechanisms by which the levels of ErbB2 protein are affected by the EBP1 (ErbB3-binding protein-1) and PAK1 levels in human breast cancer cells. Understanding this complex relationship and interplay may lead to better breast cancer therapies. Second project involved elucidating the affects of EBP1 protein binding to the ErbB2 promoter region after ligand stimulation and its affect on gene regulation. Postdoctoral fellow, Center for Vascular & Inflammatory Diseases, Jan 2007-Oct 2009 Investigated the effect of juxtacrine signalling on various signalling pathways in brain cancer cells. This work resulted in a journal publication in the Journal of Biological Chemistry in 2010. Studied the mechanism by which the interaction between the ligand, TWEAK, and its receptor, Fn14, results in invasion, metastasis and angiogenesis in brain cancers. Postdoctoral fellow, Mucosal Biology Research Center, Aug 2004-Jan 2007 Studied the effect of heat shock factor-1 (HSF-1) on cytokine gene expression under fever conditions. This work resulted in a journal publication in the American Journal of Physiology - Cell Physiology in 2010. Second project investigated which transcription factors were recruited to the 5UTR promoter site of TNFalpha during fever conditions. Understanding this process would help us better treat patients during sepsis conditions in the clinic using piperidine-cleaved DNA footprinting and ChIP. Postdoctoral fellow, Department of Pharmaceutical Sciences, April 2002-May 2004 Project involved the cloning, purification and initial structure-function characterization of ShuT (Shigella heme uptake protein) from Shigella dysenteria. Also generated new mutants for ShuT in the ligand-binding site to better understand the kinetics of hemebinding and release and generated anti-ShuT antibodies to study in-vivo expression levels in a siderophoredeficient mutant. This work resulted in a journal publication in Biochemistry in 2005.

University of London, London (UK). Ph.D. Student, University of London, Kings College, London, October 1997- April 2002. Project was concerned with elucidating the unknown structure, biochemistry and molecular biology of the secreted fungal enzyme copper-enzyme (laccase) from a leaf-litter degrading fungus M. galopus found in UK woodlands. Methods involved optimising fungal mycelium growth for enzyme production in various media and using various laccase enzyme inducers, Assaying various ligninolytic enzymes to quantify specific secreted protein like laccases, peroxidases, cellulases, mannanases and xylanases using spectroscopic and oxygen-rank electrode measurements. Developed a new technique for genomic DNA extraction from needle-litter colonised by M. galopus. Also modelled PCR-derived laccase fragment to determine amino acid/overall structural similarity of this novel laccase to other known basidiomycete laccases. This work resulted in a journal publication in Mycological Research in 2003. Radioactive assays using P32-ATP, SDS-PAGE, Western blotting, Zymograms, immunoprecipitation (IP), Co-IP, Circular Dichroism (CD), UV-Vis spectroscopy, EMSA (Electrophoretic Mobility Shift Assay), and ELISA. Molecular Biology: DNA extraction from fungal, bacterial and mammalian sources, bacterial transformation, mammalian transfections. PCR methodologies to create genomic cDNA library, gel electrophoresis, cloning, screening of phage libraries, site-directed mutagenesis, DNA footprinting, Chromatin immunoprecipitation assay (ChIP), real-time PCR (RT-PCR, qRT-PCR), Northern and Southern blotting.
