CLINITEST

PRINCIPLE
Semiquantitative method to detect the presence of glucose
and other reducing substances in feces.
Cupric sulfate is converted to cuprous oxide when copper
sulfate reacts with reducing substances.
The resultant color, which varies with the amount of
reducing substances present, ranges from blue through
green to orange.
Sodium hydroxide provides an alkaline medium necessary
for the reaction to occur. Heat required is furnished by the
reaction of sodium hydroxide with water and with citric acid.
Citric acid and sodium carbonate aid in dissolving the
tablet.

REAGENTS AND SUPPLIES


1. 16x100mm glass test tube
2. Distilled/sterile water - at room temperature
3. Beral pipette or syringe
4. Clinitest Reagent Tablets
* (contains 1 part copper sulfate, 12 parts sodium
hydroxide, 4 parts sodium carbonate, 15 parts citric
acid, filler and binder).
5. Quantimetrix Dropper Plus Urine Controls - Level 1 and
Level 2 - 5 mls per bottle. When stored at 2-8C,
unopened controls are good until expiration
date.

PROCEDURE:
A. Quality Control Testing
1. Drop 5 drops of the Level 1 control into one test tube and 5 drops
of Level 2 control into another test tube.
NOTE: If more than one specimen is being tested, the test
tube should be identified to prevent clerical errors from occurring.
Also if a pipette is used, then a pipette should be used for all steps.
If a syringe is used, then a syringe should be used for all steps.
2. Using a pipette or syringe in an upright position, drop 10 drops of
distilled or sterile water into each test tube.
3. Remove a tablet from Clinitest bottle (do not remove prior to this
time) for each tube.

4. Drop a tablet into each test tube. DO NOT touch


bottom of tube - HOT!
5. Watch while complete boiling reaction takes place.
DO NOT shake test tube during boiling, or for the
following 15 seconds after boiling has stopped.
6. At the end of the 15 second waiting period, shake
the test tube gently to mix contents.
7. Compare the color of the liquid to the Color Chart
packaged with each bottle. Ignore any sediment that
may have formed in the bottom of the tube. Also
ignore any changes after the 15 second waiting
period.

B. Patient Testing
1. Collect feces following the appropriate collection procedure.
2. In a container, place 1 volume of stool.
3. Add 2 volumes of distilled/sterile water to the container.
4. Mix thoroughly.
5. Transfer 15 drops of this mixture to a clean test tube.
6. Remove tablet from Clinitest bottle (do not remove prior to
this time).
7. Drop tablet into test tube. DO NOT touch bottom of tube
- HOT!
8. Watch while complete boiling reaction takes place. DO NOT
shake
test tube during boiling, or for the following 15 seconds after
boiling has stopped.

9. At the end of the 15 second waiting period,


shake the test tube gently to mix contents.
10. Compare the color of the liquid to the Color
Chart packaged with each bottle. Ignore any
sediment that may have formed in the bottom
of the tube. Also ignore any changes after the
15 second waiting period.
11. Document on the appropriate result log and
on the patients chart the percent (%) result
which appears on the color block that most
closely matches the color of the liquid.

REPORTING RESULTS
Expected Results:
The presence of 0.25mg/dl (1/4%) or less
is considered normal.
The presence of 0.25 to 0.50mg/dl is
considered suspicious.
The presence of 0.5.mg/dl or greater is
considered abnormal.
Color Block Designations:
Negative, 1/4% (0.25), 1/2% (0.50), 3/4%
(0.75), 1%, 2% or more.
