4 Parameter Logistic (4PL)

• F(x) = ((A-D)/(1+((x/C)^B))) + D
• Nonlinear regression
• Monotonic
– e.g. For every MFI, there is only one
concentration
• Symmetrical around the midpoint or
inflection point

> high standard OR highest extrapolated value

D = maximum asymptote

B = slope factor/steepness of curve


C = inflection point

A = minimum asymptote
< low standard OR lowest extrapolated value

Caution on Extrapolation

tiny change in MFI = HUGE change in concentration

• F(x) = A + (D/(1+(X/C)^B)^E)
• Nonlinear regression
• Monotonic
• Key difference = asymmetry
• E = asymmetry factor
• E = 1 = 4PL model equation

Assumptions of the 4PL & 5PL

• 2 assumptions about how the data are


scattered around the curve…
1. The scatter follows a Gaussian or “normal”
distribution
2. The standard deviation of the scatter (of the
response values or MFI) is the same for all
values of x (or concentrations in our case)

Weighting

• 1/Y^2 (Relative weighting) – Most commonly


used
• 1/Y – Most effective when the scatter follows a
Poisson distribution
• Mainly affects points on the lower portion of
the curve
• Test by doing before/after on the % Recovery
