 Introduction
• Electrophoresis technology is the basic
technology of molecular biology. This experiment
helps better understanding of DNA
electrophoresis tank and gel electrophoresis
technology.
 Experimental steps

• 1. First seal the two ends of the electrophoresis tank to prevent

liquid leakage after pouring plastic, insert the comb, letting the
bottom of the comb parallel to the bottom of the groove and leave a
gap. After thawing the gel (usually 1% -2%) into a clear liquid (no
lumpy gel solids), add EB. Pour the glue into the electrophoresis
tank. After the gel solidified, pull out the comb and remove the tape.
• 2. Place the electrophoresis tank in the electrophoresis apparatus,
pour the electrophoresis buffer solution in the electrophoresis
apparatus, and the buffer solution should just cover the plastic plane.
• 3. Spot in the comb slot.

• 4. Before turning on the power, pay attention to check whether the
polarity of the electrode connected to the electrophoresis tank is
consistent with the direction of the sample moving in the carrier.
Direction: the DNA sample runs from the negative electrode toward
the positive electrode. The appearance of bubbles on the electrodes
indicates that the electrophoresis started.

• 5. Observe the marker to determine the appropriate electrophoresis
time.

• 6. Wear gloves throughout the procedure. Throw away any gloved
gloves to the designated place.
 Precautions
• Electrophoresis bath should avoid contact with organic solvents
during use, because organic glass can be dissolved in organic
solvents such as hydrocarbons; do not rub the electrophoresis tank
with alcohol, for silver phenomenon will occur after plexiglass and
alcohol contact, which will affect the life of electrophoresis tank.
Wash the electrophoresis tank with detergent. Plexiglass heat-
resistant temperature is below 80 ℃, so the glue should be poured
into the gel tray when the gel solution is cooled below 80 ℃,
otherwise the comb and tray will be deformed and lead to damage.
